psychological and social factors in diet,constipation has a serious impact on the quality of life;and there is an important role in colon cancer,hepatic encephalopathy,breast disease,Alzheimer's disease and other diseases;in acute myocardial infarction,cerebrovascular disease such as constipation can lead to accidents of life;there is a close relationship between constipation and anorectal diseases,such as hemorrhoid,anal fissure.Therefore,early prevention and rational treatment of constipation will greatly reduce the serious consequences and social burden of constipation.With the improvement of modern living standard,people's expectation of quality of life is higher and higher,and more attention has been paid to some functional diseases which have been neglected.Changes in lifestyle and diet structure makes the incidence of constipation gradually increased,especially in elderly and multiparous women.Epidemiological data show that the incidence of constipation,the rate was 3%in several national survey data from the United States,there are about 4 million Americans suffer from constipation,there are 3-4 million people every year and the application of laxatives assisted defecation,constipation prevalence rate increased with age.Epidemiological data show that the incidence rate of our country is about 10%to 15%,and the elderly people over the age of 60 account for 18%to 23%.of the populationConstipation treatment drugs are more,but most of them are difficult to receive long-term efficacy,and long-term use easily lead to drug dependence and the incidence of side effects increased.Chinese medicine for constipation treatment has accumulated a wealth of experience,that functional constipation has the actual situation,the clinical discovery of intractable constipation with the actual secret based,age of onset tends to younger.Professor Zhou Jianhua,after years of practice,reference and foundation treatment of constipation,and achieved preliminary results on the development of external treatment,for long-term use of laxatives bring side effects.The effect of functional constipation is not satisfactory,because the etiology and constipation.Pro motive drugs can lead to malignant cardiovascular events and are less clinically useful.Professor Zhou Jianhua summed up the experience of treatment of functional constipation for many years,and established external treatment(constipation paste)to treat functional constipation,and the clinical efficacy was satisfactory.Zhou Shiqin for old,Huitong and western,and combining many years of clinical practice,has a unique understanding of constipation for a disease.Constipation paste is the crystallization of Professor Zhou's clinical experience for many years,and has been widely used in the anorectal area under its leadership.In order to further verify the clinical efficacy of constipation treatment with functional constipation,the mechanism of treatment and the current research progress at home and abroad are specially designed.This topic mainly from the following 2 aspects of research:Part 1 clinical researchPurpose:to verify the clinical efficacy of constipation treated with constipation paste.Methods:80 cases were collected and randomly divided into 2 groups,40 cases in treatment group and 40 cases in control group.The treatment group was given constipation paste,the control group used oral mziren capsule(active ingredient and constipation soft capsule paste effective ingredients to treat cannabis,direction).After 2 weeks of treatment,the symptom scores of two groups were compared,and further proved that the effect of constipation paste was more significant.Result:By rank sum test,the overall curative effect between groups is P=0.01<0.05,there is significant difference,so the curative effect of the treatment group is better than the control group.Group the cure and effective rate by chi square test P=0.002<0.05,illustrate the real secret reduced,and the morphological abnormalities were observed.Compared with the blank group mice,the ICC connection network was incomplete.The number of ICC in the colon of mice increased significantly after 14 days of constipation.Western medicine mosapride has certain effect on constipation mice colon ICC,but there was no statistical difference,mziren capsule can increase the number of ICC,but there are obvious differences compared with constipation and.Constipation can significantly increase the number of effective components with constipation model mice colon ICC,and there is a dose-response relationship of ICC number,ICC number of constipation with more obvious improvement,the number is almost close to the blank group mouse colon ICC.The effect of constipation paste is more obvious.The mechanism of constipation treatment with constipation is completed by ICC.Part three:the effect of constipation paste on the expression of c-kit protein in the colon of mice with constipationResults:This study intends to put the effective component from the detection of constipation intervention colonic constipation model in mice c-kit protein expression to explore mechanism of constipation paste,so as to explain its cathartic mechanism at the molecular level.Methods:the mice models of constipation were treated by liquid nitrogen cryopreservation,and the expression of c-kit protein in the colon tissues of mice with constipation induced by constipation was detected by Western-blot technique.Results:polyacrylamide gel electrophoresis and Western blot of nitrocellulose membrane showed a band of 145KD,which was consistent with the molecular weight of c-kit protein.The expression of the protein in the model group was significantly decreased,and the expression of the protein in the constipation paste group was significantly increased compared with the model group.By means of analysis of variance,there was significant difference between the model group and the blank group in P<0.01,indicating that the expression of c-kit protein in the colon of the model group mice was obviously down regulated,and there was an abnormal expression of the
