Purpose:In this paper,through the preparation of middle cerebral artery occlusion reperfusion(MCAO)model in rats,using electric acupuncture at Baihui,Yintang and double Zusanli points,to clarify that electroacupuncture can promote the expression of PRG5 protein,inhibit the expression of Rho A protein,reduce the damage of nerve function,and repair the nerve function,provide a new theoretical basis for the clinical application of electroacupuncture in the treatment of cerebral ischemia According to.Material and method:Thirty healthy male SD rats,10-12 weeks old,weighing 300 ±40g,were fed adaptively for one week before the experiment.The method of random number table was divided into two groups: sham operation group(6 rats)and model replication group(24 rats).According to the method of Zea longa thread embolism,the model of middle cerebral artery occlusion reperfusion injury was made in rats.In the sham operation group,only the artery was dissected and no embolization was performed.After the model was established,the rats were graded according to the longa scoring standard(excluding the dead rats and the rats with 0 and 4 scores),and the rats in the model replication group were randomly divided into the control group and the electroacupuncture group.The rats in the electroacupuncture group were treated with Baihui,Yintang and Zusanli points respectively.The rats in the control group and the sham operation group were only bound for the same time without electroacupuncture.Every time 10 minutes,every 24 hours,for 2 weeks.At the end of the intervention,the rats were scored for neurological function again.After the score,the adhesive removal experiment was carried out.Finally,all the rats were killed and the brain tissues were taken out.The expression of PRG5 and Rho A protein in the ischemic brain tissues was detected by Western blotting,immunohistochemistry and immunofluorescence.Spss19.0 software was used for statistical analysis.Results:1.Neurological function score: after modeling,the neurological function score of the sham operation group was 0,with no neurological function injury.The neurological function score of the model replication group was significantly higher than that of the sham group,with statistical significance(P < 0.01).After 2 weeks of intervention,compared with the sham group,the neurological function score of the electroacupuncture group was significantly higher than that of the sham group(P < 0.01).Compared with the control group,the neurological function score of electroacupuncture group was significantly lower than that of control group,with statistical significance(P < 0.01).2.Adhesive removal experiment: after 2 weeks of electroacupuncture intervention,the adhesive removal experiment results showed that compared with the sham operation group,the time spent in the electroacupuncture group was significantly longer than that in the sham operation group,with statistical significance(P < 0.01).Compared with the control group,the time spent in the electroacupuncture group was significantly lower than that in the control group,with statistical significance(P < 0.01).3.Western blot detection: compared with the sham operation group,the expression level of PRG5 protein in ischemic brain tissues of rats in the electroacupuncture group was significantly decreased,and the expression level of Rho A protein was significantly up-regulated(P < 0.01).Compared with the control group,the expression level of PRG5 protein was up-regulated and the expression level of Rho A protein was decreased(P < 0.01).4.Immunohistochemical detection: compared with the sham operation group,the number of PRG5 positive cells in the ischemic brain tissue of rats in the electroacupuncture group was significantly decreased,and the number of Rho A positive cells was significantly up-regulated(P < 0.01).Compared with the control group,the number of PRG5 positive cells was significantly up-regulated and the number of Rho A positive cells was significantly decreased in the electroacupuncture group(P < 0.01).5.Immunofluorescence detection: compared with the sham operation group,the average optical density of PRG5 in ischemic brain tissues of the electroacupuncture group decreased significantly,while the average optical density of Rho A increased significantly(P < 0.01).Compared with the control group,the average optical density of PRG5 in ischemic brain tissues of the electroacupuncture group was significantly increased,while the average optical density of Rho A was significantly decreased(P < 0.01).Conclusion:1.Electroacupuncture at Baihui,Yintang and Zusanli points can significantly improve the neurological deficit of rats with cerebral ischemia-reperfusion injury and protect the cerebral ischemia tissue.2.Electroacupuncture at Baihui,Yintang and Zusanli points can improve the motor function of rats with cerebral ischemia-reperfusion.3.Electroacupuncture at Baihui,Yintang and Zusanli points can increase the expression of PRG5 protein and decrease the expression of Rho A protein in ischemic brain tissue,reduce the damage of nerve function after stroke,and promote the repair of nerve function.
