Purpose:In this study,a rat model of type 2 diabetic nephropathy induced by high-sugar and high-fat diet combined with STZ injection was established to detect the blood glucose,blood creatinine(Sur),urea nitrogen(BUN)and renal weight/body weight of the model rats,and to discuss the scientific basis of internal and external treatment of Chinese medicine on model rats to prevent and delay diabetic nephropathy.The contents of transforming factor 1(TGF-1)and type IV collagen(col-iv)in the renal tissues of the model rats were detected to explore the mechanism of the effect of internal and external treatment of Chinese medicine on the renal damage in model rats.This experiment is based on the theory of "the principle of internal treatment is the principle of external treatment" to explore the principles of prevention and treatment of diabetic nephropathy,in order to provide experimental evidence and theoretical guidance for the clinical diagnosis and treatment of diabetic nephropathyMaterial and method:Male Wistar rats were weighed and numbered,and 50 rats were randomly selected as blank control groups using random number table methods,and fed with ordinary feed.The remaining 40 rats were fed with high-sugar and high-fat feeds,and fed 4 weeks,the body weight reached 340g ± 10g,and 4 of them did not meet the standard and were excluded from the experiment.After fasting for 12 hours,intraperitoneal injection was performed at a dose of 50 mg/kg.After 72 hours,fasting blood glucose was measured?16.7 mmol/L and included in the observation subject.If the blood glucose was not up to standard,STZ drugs were replenished once.Sterile citrate buffer solution,in which 3 rats with hypoglycemia were removed from the experiment,the blank control group was fed with ordinary feed,and the remaining four groups continued to be fed with high-sugar and high-fat feed and continued to feed for 12 weeks.8 rats were randomly selected from the blank control group and the remaining models,and blood was taken from the orbital vein.The biochemical automatic analyzer method was used to quantitatively detect Sur and BUN for statistical analysis.If the values of the two groups were P<0.05,it was statistically significant,indicating that the rats had renal damage.During the modeling of diabetic renal damage,5 rats died due to intolerance.Rats in the model group were randomly divided into model control group,internal treatment group,external treatment group,and moxibustion combination group,with 7 rats in each group.The internal treatment group was given jinguishenqi pill and wuziyanzong pill decoction plus or minus gavage treatment(4ml/time,1 time/d).The external treatment method used aconite,cinnamon and other drugs to grind rice wine and mix and apply externally to Shenque points,1 time/d;The moxibustion combination group was given a combination of internal and external treatments;the blank control group and the model control group were given the same amount of distilled water for gastric administration.The above treatment cycle was 4 weeks.During the treatment,the rats in each group were fed according to the established manner.After treatment,blood glucose,creatinine,urea nitrogen and renal weight/body weight of rats were measured,and TGF-TGF 1 and COL-IV in renal tissue were detected by Western BlotResults:1.After treatment,the general state of the model rats,such as less movement and crouching,dark fur,and frequent urination,was improved compared with that before treatment2.Blood glucose:after treatment,the blood glucose value of the model control group increased significantly.Although the blood glucose value of each treatment group was higher than that of the blank control group,it showed a downward trend compared with that of the model control group,showing a statistical difference(P<0.01)3.Renal index of rats(renal weight/body weight value):the renal index of the model control group was compared with that of the blank control group(P<0.05),showing a statistical difference,indicating that the kidneys of rats have hypertrophic pathological changes.The renal index of each treatment group was compared with that of the model control group,P<0.05,and the difference was statistically significant,indicating that the various treatment methods had certain effect.There was no significant difference in renal index between the internal treatment group and the external treatment group(P>,0.05),but the renal index ratio of the moxibustion-medicine combination group was significantly better than that of the internal treatment group and the external treatment group(P<0.05)4.Renal function(blood creatinine Sur,urea nitrogen BUN):the values of Sur and BUN of the model control group were significantly higher than those of the blank control group,it indicates that the renal land the renal function of the combined moxibustion group was significantly better than that of the other four groups(P<0.05),while there was no statistical difference between the internal treatment group and the external treatment group(P>0.05)5.Western-blot detection of TGF-1 and col-iv in renal tissues:Compared with the blank control group,the expression levels of TGF-?1 and COLIV in the model control group were increased,and the gray values were statistically different(P<0.01);the expression levels of TGF-1 and col-iv in each treatment group were significantly decreased compared with the model control group(P<0.05),showing statistical differences,indicating that the various treatment methods had certain effects.There was no significant difference in the expression levels of TGF-1 and col-iv factors in the internal and external treatment groups(P>,0.05),however,compared with the internal treatment group and the external treatment group,the expression levels of TGF-?1 and COL-IV factors in the moxibustion-medicine combination group were significantly reduced(P<0.05),indicating that the kidney function of the moxibustion-medicine combination group was significantly better than that of the internal treatment group and the external treatment groupConclusion:1.The animal model of early renal damage in type 2 DM can be accurately reproduced by feeding with high-sugar and high-fat diet combined with certain dose of STZ intra-abdominal injection,which is an ideal model for studying DN in traditional Chinese medicine2.The combination of internal and external treatment can change the general state of the model rats and improve certain clinical symptoms3.Combination of moxibustion drugs can reduce the levels of blood glucose,kidney weight/weight,BUN and Sur,and effectively delay the development of DN4.Combination of moxibustion drugs can reduce or downregulate the content of TGF-TGF-IV 1 protein and COL-IV,effectively inhibit the process of renal fibrosis,and achieve the effect of prevention and treatment of DN...
