OBJECTIVE: The new wound oil agent of sichuan prickly ash is a new medicine of orthopaedic hospital of sichuan province,which is used to treat the early rapid anti-inflammatory,detumescence and analgesia of closed fracture.This study through the establishment of closed-loop tibiofibula fracture model,to observe Chinese prickly ash new injury of agents in the soft tissue injuries of SD rats symptomology,histomorphology and organization of IL-1?,TNF-?,PGE2,COX-2,mPGES-1 changes in the indexes of mRNA,which discuss Chinese prickly ash new anti-inflammatory mechanism of injury of agents for the treatment of early fracture damage,in order to provide a basis for pharmacology in new drug research and development,and also lay the foundation for late clinical promotion.Methods: Seventy-two eight-week-old SD male rats were randomly divided into a blank group and a model group according to the random number method.According to the time points of the experimental design,each group was further divided into 1 day group,4 day group and 7 day group,with 6 mice in each group.Double fractures of the right tibia and fibula bone in the model group were caused by the self-made closed fracture molding device.Every day in rats with bilateral leg skin temperature change and the observation,record the limb injury symptom index,the other in the building after 1 day,the fourth day,7 days of each group rats on the right side of the posterior tibial muscle tissue based,for then the histologic morphology observation,ELISA detection organization IL-1?,TNF-?,content of PGE2 and PCR to detect COX-2 mRNA,m PGES-1 mRNA expression.Results:(1)Skin temperature: both the new wound oil agent group and the futaolin group could reduce the skin temperature difference,which was significantly different from the normal saline group and the blank group(P<0.05).(2)Injury symptom index: both the oil agent group and the futaolin group could improve the symptoms and signs of soft tissue injury,which were significantly different from the normal saline group and the blank group(P<0.05).(3)Histopathology: compared with the normal saline group,both the new wound oil agent and futalin could inhibit the invasion of inflammatory cells and promote the repair of tissues.(4)ELISA:IL-1?,TNF-? and PGE2 in the muscle tissue of the model group were significantly higher than those of the blank group.IL-1? 1 day,model group,the content of TNF-? and PGE2 was no obvious difference(P > 0.05),Chinese prickly ash new injury of agents and helped him Lin groups on the fourth day,7 days can significantly reduce the IL-1?,TNF-? and the content of PGE2 compared with lifted him Lin group,in terms of inhibition of IL-1?,there was no significant difference(P > 0.05),the inhibition of TNF-? and PGE2 in 4 days,there are significant differences(P < 0.05),and 7 day have no obvious difference(P > 0.05);Moreover,there were significant differences between the new wound oil agent group and futaolin group and the normal saline group and the blank group(P<0.05).(5)PCR detection: the expressions of COX-2 mRNA and mPGES-1 mRNA in the tissues of the model group were significantly higher than those of the blank group at three time nodes.Day 1,the model group the three groups of COX-2,mPGES-1 are in high the mRNA expression,no significant difference(P > 0.05),Chinese prickly ash new injury of agents and helped him Lin groups in the fourth day,7 days after building can cut IL-1?,TNF-?,and the content of PGE2 compared with lifted him Lin group,in the lower COX-2 mRNA expression,there was no significant difference(P > 0.05),on the fourth day cut mPGES-1 mRNA expression,with significant difference(P < 0.05),and 7 day have no obvious difference(P > 0.05);Moreover,there were significant differences between the new wound oil agent group and futaolin group and the normal saline group and the blank group(P<0.05).Conclusion:(1)The new wound oil agent of sichuan peppercorns can effectively improve the symptoms,signs and tissue morphology of the early fracture soft tissue injury,and has an exact effect on tissue repair.(2)Chinese prickly ash new injury of agents for the treatment of early fracture damage have definite anti-inflammatory effect,its mechanism may be through lower IL-1? in the local tissue injury,the content of TNF-? and lower COX-2 mRNA,m PGES-1m RNA expression in order to reduce the synthesis of PGE2,thus reduce the inflammatory response,achieve the goal of fast anti-inflammation,detumescence and analgesic,and help the organization quick fixes.
