The prevention and treatment of malignant tumors should not be ignored.Tumor blood vessels and tumor microenvironment,as important factors of tumor growth and metastasis,are also paid more attention.Professor Huang Jinchang has rich experience in the treatment of tumor,especially in the treatment of peritumoral acupuncture.Peritumoral acupuncture,as a multi needle encircling acupuncture method,is effective in the treatment of tumors.The results of our team's previous experiments indicate that the peritumoral acupuncture may be through VEGF to inhibit tumor angiogenesis,which result to the effect of tumor inhibition.However,the mechanism of inhibition of angiogenesis by peritumoral acupuncture via VEGF pathway is not clear.Therefore,the purpose of the study is to try to make it clear.This study combines a total of two parts(theoretical discussion and experimental study)in addition to a review.The review describes VEGF and VEGFR,with particular focus on their role in angiogenesis.The theoretical discussion gives an account of Professor Huang Jinchang's experience in the treatment of tumor with the theory of "obstruction of tumor collaterals stasis".The experimental study is applied to study the mechanism of the inhibition of angiogenesis by peritumoral acupuncture through VEGF pathwayThe theoretical discussion part of this study summarizes Professor Huang Jinchang's experience in the treatment of tumor with the theory of " obstruction of tumor collaterals stasis ".Professor Huang Jinchang proposed the theory of " obstruction of tumor collaterals stasis" as the core pathogenesis of tumor based on the collateral disease theory,with tumor blood vessel as the structural basis and tumor microenvironment as the biological basis Tumor belongs to the category of collateral disease." Obstruction of tumor collaterals stasis"is not only the core pathogenesis of tumor,but also the important factor of tumor growth and development.Professor Huang Jinchang guided the clinical treatment of tumor with this theory.According to the treatment principle of "unobstructed collaterals are available",he used different methods to treat tumor and tumor complications,such as peritumoral acupuncture,blood-letting puncture,moxibustion and traditional chinese medicine.Peritumoral acupuncture can directly act on the peritumoral area and is the main method to improve the obstruction of tumor collateral stasis.In this study,immunohistochemistry was used to observe the angiogenesis related indexes such as CD34,VEGF-A,VEGFR-2,PI3K,Akt,Ras and MAPK in 4T1 breast cancer mice after elctroacupuncture to investigate the mechanism of angiogenesis inhibited by peritumoral acupuncture through VEGF pathway.Objective:To investigate the mechanism of angiogenesis inhibited by peritumoral acupuncture through VEGF pathway.Methods:This is an animal experiment of 4T1 breast cancer mice transplanted tumor model.After three days of peritumoral acupuncture,the CD34,VEGF-A,VEGFR-2,PI3K,Akt,Ras,MAPK indexes were observed to clarify the mechanism of the inhibition of angiogenesis by elctroacupuncture through VEGF pathway.Result:1.4T1 breast cancer transplantation model and general situation evaluation:the the survival status of mice and the growth state of tumor in each group is well.There was no significant difference in body weight and tumor volume between the groups at T0 time point,and the baseline was uniform and comparable.2.CD34 expression:compared with TG,the CD34 expression of EATG and BTG decreased,and the difference was statistically significant.3.VEGF-A expression:compared with TG,the VEGF-A expression of EATG and BTG decreased,and the difference was statistically significant.4.PI3K expression:compared with TG,the PI3K expression of EATG and BTG decreased,and the difference was statistically significant.5.Akt expression:compared with TG,the Akt expression of EATG and BTG decreased,and the difference was statistically significant.6.Expression of VEGFR-2,RAS and MAPK:there was no significant difference in the expression of VEGFR-2,RAS and MAPK in each groupConclusion:the inhibition of angiogenesis of 4T1 breast cancer mice by peritumoral acupuncture may be achieved by VEGF-PI3K-Akt pathway.
