Objective:Using ligation of LAD combined with STZ intraperitoneal injection to establish the model of coronary heart disease combined with hyperglycemia in rats,and making a comprehensive and dynamic observation.The syndrome and time window of the model were determined by collecting the macroscopic and microscopic indexes of rats.On this basis,further explore the biological characteristics of the syndrome model.Method:1.After 7 days of adaptive feeding,male SD rats were randomly divided into sham operation group,high glucose group,ligation group and high glucose ligation group.High glucose group was given one-time intraperitoneal injection of STZ(55mg/kg),ligation group was given lad ligation,high glucose ligation group was given one-time intraperitoneal injection of STZ(55mg/kg)after lad operation.At 7,14 and 28 days after operation,ECG,blood glucose,tension test,respiratory rate of stress state,RGB value of foot image,amount of drinking water,body mass,echocardiography,heart and liver pathology were collected to determine the success of the model and its syndrome attributes.2.The indexes of blood central muscle injury(AST,CK,CK-MB,LDH),blood lipid(TG,HDL-C,LDL-C,CHOL),inflammatory factor(TNF-?,IL-1?,IL-6),blood coagulation(PT,APTT,t t,FBG,fbg-t),fibrosis(CVF)were detected,and the correlation between them and syndrome indexes was analyzed.Result:1.In the high glucose ligation group,pathological Q wave or S-T segment elevation appeared in ECG,and fasting blood glucose?16.7mmol/l.Compared with the sham operation group,the EF and FS values of the patients in the 7-28 days after operation were significantly reduced(P<0.01),and the cardiac muscle structure was disordered 28 days after operation,which proved that the model of coronary heart disease combined with hyperglycemia was established successfully.Compared with the sham operation group,the rats in the model group had a significant decrease in the tension value(P<0.01),a significant increase in the respiratory frequency under stress(P<0.05),an increase in the amount of food and water(P<0.05),a significant decrease in body weight,and macroscopic signs of dry stool and yellow urine,which were consistent with the syndrome of deficiency of Qi and Yin.At 7-28 days,the R ratio of tongue and foot decreased significantly(P<0.01),the number of small vessels on the back of ear increased,and the color of the tissue below the ligation line became darker,which was in accordance with the syndrome of blood stasis.2.Compared with the sham operation group,LDH and CK-MB in the model of high glucose ligation group were significantly increased in 7-28 days(P<0.01),AST was significantly different in the whole time point(P<0.01),CK was significantly different in 7-28 days(P<0.05),and there was no difference in 14 days.In general,myocardial injury almost throughout the observation period.Compared with sham operation group,TG and LDL-C in model group increased significantly in 7-28 days(P<0.01),HDL-C in 7-14 days was lower than sham operation group(P<0.05),but there was no difference in 28 days.The levels of TNF-?,IL-1? and IL-6 were significantly higher in the high glucose ligation group than in the ligation group(P<0.05).Blood coagulation abnormality was accompanied by the whole pathological process of the model group,and the value of FBG was significantly higher than that of the sham operation group(P<0.01).Myocardial fibrosis was serious in high glucose ligation group,and CVF value increased significantly with time(P<0.01).The results of correlation analysis showed that 6 and 5 indexes were involved in the formation of Qi Yin deficiency syndrome and blood stasis syndrome,and there was a cross.Conclusion:1.Hyperglycemia combined with LAD ligation induced coronary heart disease combined with hyperglycemia model belongs to Qi Yin deficiency and blood stasis syndrome 7-28 days after operation.2.During the whole process of the model,there were myocardial injury,lipid metabolism abnormality,coagulation abnormality,inflammatory reaction,myocardial fibrosis,and more progressive aggravation.6 and 5indexes participated in the formation of Qi Yin deficiency syndrome and blood stasis syndrome respectively,and there was a cross between them.
