A refinement to the semi-quantitative RT-PCR method and defense related, stress resistance and insulin pathway gene expression during Sarcophaga crassipalpis diapause

Abstract/Summary:
To screen for differential gene expression between flesh fly diapause and post-diapause states, total RNA from diapause and early broken diapause flies was submitted for analysis using a Drosophila microarray. Genes showing greater than 5 fold up- or down-regulation in diapause were tested using degenerate primers with semi-quantitative RT-PCR and specific primers with real-time PCR. A new gene, IAP-primed, was found to be significantly up-regulated during diapause. Tenascin , a pair-rule gene, was found to be significantly down-regulated during diapause. Insulin-like peptide 4 (Ilp4) was identified by the microarray as down-regulated but an investigation of downstream insulin pathway genes: chico, PTEN, Akt, Foxo, and 4ebp failed to find diapause related regulation.
