Glycosylation catalyzed by UDP-dependent glycosyltransferases(UGTs)is one of the key post-modifications in plants.The plant UGTs,rendering a various of secondary metabolites and plant hormones,not only play important roles for plants adapting to different environments,but also endow plant natural products with great pharmaceutical and ecological significance.In recent years,it has received extensive attention with the increasing number of characterized plant UGTs in the biosynthesis of many bioactive compounds.To date,thousands of plants genomes and transcriptomes have been sequenced,but only 10% of plants contained UGT gene annotations.It severely hampered the functional characterization,gene mining and engineering of important UGTs.Here,we provide a comprehensive plant UGTs resource by constructing a plant UGTs database(p UGTdb)(http://pugtdb.biodesign.ac.cn/),which integrated the UGTs mainly from many data sources.First,a total of 143,941 UGTs were collected from 799 plant genomes in NCBI by integrating NCBI annotation(31,886)and GMind annotation(112,055);Second,8,004 UGTs were collected from other 57 public genomes which were not submitted to NCBI;Third,60,622 UGTs were collected from 1,087 plant transcriptomes assembled in 1KP database.In total,there are 212,567 UGTs from 1,943 plants collected in p UGTdb.Furthermore,all plant UGTs in p UGTdb were further divided into 331 UGT families and 10,070 UGT subfamilies.Among them,there are 278 new UGT families,which have no formal nomenclature in previous studies.Finally,we developed a tool for the functional prediction of plant UGTs by analyzing the sequence and structure of sugar donors and acceptors in 294 known plant UGTs.In addition,by using this functional prediction tool,we successfully mined the key UGT gene involved in the biosynthesis of polydatin.In summary,this work not only develops useful tools for gene annotation and function annotation for plant UGTs,but also provides a comprehensive platform for gene mining,functional identification,and biosynthesis of important plant natural products.
