Using ultraviolet-visible spectropHotometer method determinate the content of glyceride in massoniana pulp.By glyceride product color full wavelength scanning experiments,got the glyceride product color for maximum absorption peak at 410 nm.Recovery rate and precision experiments validate the method had higher accuracy and operability.Using the method of the determination of glyceride,draw the glyceride product color standard curve and the equation for Y=4.5626+0.1186x(R2=0.9872),By ultraviolet-visible spectropHotometer determinate the absorbance values,Measured in pinus massoniana wood pulp samples glyceride concentration is 0.01599 g/g(dry pulp).Through single factor experiments of lipase treatment of masson pine wood pulp resin,the optimum level of lipase dosage was 20U/g,the best value of pH was 7,the best value of reaction temperature was 60?,the plasma concentration of optimal value was 6%,the best value of reaction time was 2 h.Based on the single factor determine the best level of above,using the response surface analysis for the handling of the resin in pinus massoniana wood pulp by lipase to optimize the reaction conditions.Four factors on the influence degree of the lipase in pinus massoniana wood pulp resin in the order:Pulp thick>pH>enzyme dosage>reaction temperature.Lipase function optimization conditions of pinus massoniana wood pulp was:the best value of enzyme dosage was 18.90 U/g,the best value of plasma concentration was 5.5%,the optimal value of reaction temperature was 62.8,the best value of pH was 7.63.the best value of reaction time was 2h.Under the optimal conditions,the degradation rate of lipase hydrolyzsis glyceride of pinus massoniana wood pulp was 16.1%.Through the Orthogonal test of laccase hydrolysis glyceride in massoniana pulp,the optimum reaction conditions of laccase hydrolysis glyceride in massoniana pulp was:Enzyme dosage was 2%,the pulp concentration was 4%,pH value of 6,reaction temperature of 40?.Comprehensive consideration of the optimum reaction condition of lipase and laccase,selected massoniana pulp concentration was 5%(m),respectively,the optimal reaction conditions in their respective processing massoniana wood pulp,the reaction time for 1h.Through the experimental group and control group of experiment data to calculate the laccase combined with lipase hydrolysis glyceride in massoniana pulp degradation rate was 19.4%.Lipase combined with surfactant hydrolysis glyceride in pinus massoniana experimental results showed that:among the selected several kinds of surfactants,the song X-100,twain-80,polyethylene glycol-400,polyethylene glycol-6000,sodium dodecyl sulfate can promote lipase hydrolyzed glyceride in pinus massoniana wood pulp.In the optimization of five kinds of surface active dosage of single factor experiment,the most suitable amount of Song X-100 was 1.24%(oven dry pulp),the most suitable amount of twain-80 was 1%(dry pulp),the most suitable amount of polyethylene glycol-6000 was 0.52%(dry pulp),the most suitable amount of polyethylene glycol-400 was 1%(dry pulp),sodium dodecyl sulfate is the most suitable amount of 0.27%(dry pulp).The experiment of lipase combination with the optimizing surfactant hydrolysis glyceride in the pulp,the degradation rate of lipase combined with polyethylene glycol-400 hydrolysis glyceride in pinus massoniana was 21.3%,the degradation rate of lipase combined with songX-100 hydrolysis glyceride in pinus massoniana was 24.7%,the degradation rate of lipase combined with twain-80 hydrolysis glyceride in pinus massoniana was 19.0%,the degradation rate of lipase combined with sodium dodecyl sulfate hydrolysis glyceride in pinus massoniana was the highest,the degradation rate reached 28.7%.Comprehensive comparison:the degradation of lipase hydrolyzed glyceride of pinus massoniana wood pulp at a rate of 16.1%,laccase combined with lipase hydrolysis glyceride in massoniana pulp degradation rate was 19.4%.the degradation rate of lipase combined with sodium dodecyl sulfate hydrolysis glyceride in pinus massoniana was 28.7%.Finally,it is concluded that lipase combined with surfactant hydrolysis glyceride in pinus massoniana was the best way.
