In the paper,the chemical compositions analysis and the animal behavior test were combined to compare the hypnotic effects of Rosa setate×Rosa rugosa essential oil(RREO)and Rosa damascena essential oil(RDEO),in which RREO was used as the main material,and the mechanism of hypnotic effect of RREO was studied by neurochemical experiment.The main conclusions were as follows:Chemical composition analysis of RREO:56 kinds of chemical substances were detected from RREO,mainly consisting of alcohols(63.61%),olefins(7.54%),alkanes(5.01%),aldehydes(4.77%),ketones(3.30%),ethers(1.49%)and other chemicals,the highest components of RREO were citronellol(41.49%)and geraniol(11.06%).49 kinds of chemical substances were detected from RDEO,mainly consisting of alcohols(56.53%),alkanes(28.29%),alkenes(9.19%),esters(1.66%),ethers(1.05%)and other chemicals,the highest components of RDEO were citronellol(35.31%)and geraniol(17.26%).Animal behavioral test:spontaneous activity,pentobarbital sodium-induced sleep time and body temperature are often used to determine whether the test drugs have hypnotic effects or not.The results showed that both RREO and RDEO had hypnotic effects,and the optimum concentration of RREO(1×10-3g/ml)was better than that of RDEO(1×10-3g/ml),as RREO could significantly shorten the sleep latency of ICR mice,compared with the control group,prolong sleep duration,reduced the amount of autonomic activity,and significantly lower body temperature in mice.Animal behavior experiments were carried out with the absolute concentration of citronella and geraniol in the optimum concentration of bitter water rose essential oil,and the experimental results showed that:citronellol also showed hypnotic effect in behavioral tests,but its effect was inferior to that of essential oil,and the sleep duration induced by citronellol was significantly lower than that of essential oil,in addation,the citronellol did not cause significant changes in body temperature of mice.The bioactivity of chiral compounds is related to its configuration.To determine the relationship between the bioactivity and the stereoconfiguration of citronellol,the three kinds of citronellol with different stereoconfiguration were evaluated for hypnotic effects.The results showed that L-citronellol had a hypnotic effect,(±)-citronellol could prolong the duration of sleep,but could not reduce the amount of spontaneous activity of mice,D-citronellol did not show sedative-hypnotic effects.Neurochemical experiment:The mice were injected with PCPA to make the insomnia model,and the neurotransmitters were detected by HPLC-ECD.The experimental results showed that the PCPA insomnia model reduced the content of 5-HT and 5-HIAA in the brain of mice,and increased the level of NE and DA in the hypothalamus.Compared with the insomnia model,RREO significantly increased the 5-HT in the hypothalamus,hippocampus and striatum of mice and significantly increased the 5-HIAA in the hypothalamus,hippocampus,striatum and cerebral cortex of mice,significantly reduced the DA in the hypothalamus and hippocampus and accelerated its metabolism,significantly reduced the NE in the hypothalamus;L-citronellol significantly increased the 5-HT in the hypothalamus of mice,significantly increased the HVA in the cerebral cortex.In addition,both RREO and L-citronellol significantly increased the level of DA and its metabolite HVA in the striatum.
