With the rapid development of Chinese wine industry and the rapid growth of the consumer market,the fake sale of wine seriously threatens the sustainable and healthy development of the industry.Therefore,it is of great practical value to prevent and counterfeit wine by establishing an identity fingerprint information index system based on the authenticity of wine.In order to explore and screen the identity fingerprint information of wine,the carbon stable isotopes of ethanol and glycerol,oxygen stable isotopes of water and16 mineral elements in wine were selected as the pre-selection indicators of wine identity fingerprint information based on the analysis methods and related studies of stable isotopes and mineral elements.Taking the Cabernet Sauvignon dry red wine of Huaxia Great Wall Wine Co.,Ltd.of Changli County in 2017 as the test material,the selected indexes of wine were determined in 10 consecutive cycles of a month cycle according to five different bottle-sealing methods to verify the stability of the selected index system during wine storage.In this study,the determination methods of target stable isotopes and mineral elements were optimized and validated.Based on the study of the variation of stable isotopes and mineral elements in wine under different storage time and different bottle sealing methods,the identification fingerprint information index system with good stability in selected wine samples was preliminarily screened out.The main research contents and results are as follows:(1)The carbon stable isotopes of ethanol and glycerol in the wine samples were stable during storage.The carbon stable isotope ratios(?13C value)of ethanol and glycerol in 150bottles of wine samples by different bottle-sealing methods in 10 cycles were determined by liquid chromatography-stable isotope ratio mass spectrometry(LC-IRMS).The method was optimized by using 5 and 20?g/L as the injection volumes of ethanol and glycerol respectively.The results of method validation showed that the method was accurate and effective for the determination of samples.The range of?13C values of ethanol and glycerol in all samples were-28.93‰to-26.04‰and-33.48‰to-31.30‰with an average of-27.37‰and-32.18‰respectively.The average value of?13C of ethanol and glycerol was not significantly affected with the increase of storage time in different periods.There were significant differences in the average value of?13C of ethanol in different bottle-sealing methods,showing the trend of natural plug<polymerization and high polymer plug<“1+1”plug and spiral cap,and no effect on the average value of?13C of glycerol in wine samples.(2)The oxygen stable isotope of water in the wine samples were stable during storage.The oxygen stable isotope ratios(?18O value)of water in 150 bottles of wine samples by different bottle-sealing methods in 10 cycles were determined by Aqua-stable isotope ratio mass spectrometry(Aqua-IRMS).Based on the principle of carbon dioxide-water oxygen stable isotope equilibrium exchange,RM 8536 is taken as the reference standard of oxygen stable isotope to ensure the accuracy and effectiveness of sample determination.The range of?18O values of water in all samples were 1.34‰to 3.69‰with an average value of2.19‰.There was a partial significant difference in the average value of?18O of water in different periods,but it was not directly related to the storage time.The average value of?18O of water was at the same level among different bottle-sealing methods,and there was no significant difference in the average value of?18O of water between different bottle-sealing methods.(3)The four mineral elements,Co,Cr,Fe and Zn,were stable during storage in the wine samples.The mass concentration of 16 mineral elements in 150 bottles of wine samples by different bottle-sealing methods in 10 cycles were determined by inductively coupled plasma mass spectrometer(ICP-MS).The standard curve,detection limit and recovery of the method were validated.The results showed that the method could meet the needs of sample determination.There were significant differences in the average mass concentration of 16mineral elements in all wine samples during different periods,among which Co,Cr,Fe and Zn had a little significant difference.The average mass concentration of Co,Cr,Fe and Zn in all wine samples was 4.56,49.0,2051 and 686 ug/L respectively,and the change trend of mass concentration maintained the level with the increase of storage time.There was no significant difference in the average mass concentration of Co,Cr,Fe and Zn among different bottle-sealing methods.
