Effect Of Lactobacillus Plantarum Fermented Apple Juice On The Antibiotic-induced Gut Dysbacteriosis Of Mice

[Objective] This study was to investigate the alleviation effect and mechanism of antibiotic-induced dysbacteriosis in apple juice before and after fermenting,and provided a theoretical basis for improving intestinal flora imbalance.[Methods] Forty-five ICR male mice(3-week-old)were randomly divided into 5 groups.The mice in all groups were administered twice a day,for 7 days.All mice were weighed at 9 am,then the control group was given 0.3m L of sterile physiological saline by gavage,the other four groups were given an equal amount of 4 g/(kg ·d)Ceftriaxone sodium solution.After 4 hours,the mice in control group and model group were given 0.3m L of sterile physiological saline,the same amount of CAJ and FAJ were given to the CAJ group and FAJ group,respectively.The mice in LP group were given the same amount of MRS fermentation broth.During the experiment,the body weight,fecal output weight in 2h,total number of fecal output and normal recovery were observed and recorded daily.On the eighth day,the mice were sacrificed and collected the liver,kidney and spleen to measure organ ratio,and fresh feces were collected for high-throughput sequencing of 16 S r RNA to detect changes in intestinal flora.[Results] 1.The results of single factor experiment showed that the p H value of FAJ was significantly lower than that of CAJ(P<0.05);The phenolic content and tannic acid concentration increased significantly(P<0.05);the concentration of lactic acid bacteria increased significantly compared with MRS bacterial solution(P<0.05).2.The optimal ferment conditions of FAJ was: fermentation time 23 h,inoculum amount 1%,p H7.0.3.Seven consecutive days of modeling results showed:(1)Compared with the control group,the water content of feces and fecal output weight in 2h increased significantly(P<0.05)of 4g/(kg ·d),8g/(kg ·d)ceftriaxone sodium;and the water content of 4g/(kg ·d)ciprofloxacin was significantly increased(P<0.05);the numbers of Lactobacillus,Bifidobacterium,Enterococcus and Escherichia coli in the feces of the four model groups were significantly decreased(P<0.05).4.Relief test was performed on mice with intestinal flora imbalance,and the results showed:Compared with the model group,the fecal weight of 2h and the total number of fecal in the control group decreased significantly(P<0.05)from the day 2,and the other three groups began to show significant decrease on day3(P<0.05);the CAJ group showed a significant increase on the day 3(P<0.05);after day 4,there was a significant decrease in the three test groups(P<0.05);compared with the model group,on the day4,the stool rate of the control group and the three test group mice decreased significantly(P<0.05);Compared with the model group,FAJ increased the content of lactobacilli in the intestine and reduced the proportion of enterococci,thereby gradually balancing the intestinal homeostasis.[Conclusion] 1.Lactobacillus plantarum can grow normally in AJ,and the concentration of flavor components and lactic acid bacteria increased significantly after fermentation;2.Both ceftriaxone sodium and ciprofloxacin can be used to induce the establishment of a model of intestinal flora imbalance in mice;3.Compared with CAJ and probiotics,FAJ has a better effect on the intestinal flora imbalance in mice.
