Paeonia suffruticosa Andr.belongs to the perennial woody plant in the family Paeoniaceae,and had certain edible and medicinal value.As a by-product of peony peel production,peony seeds were rich in protein in addition to a variety of unsaturated fatty acids such as linoleic acid.After defated peony seeds,the protein content in seed meal can reach 20-30%.Peony seeded meal after peony seeded oil extraction was often used for feed or livestock and poultry breeding,and its deep processing and utilization are less.Therefore,it is of practical significance to study the further application of protein in peony seed meal.In this study,peony seeded protein was used as the main object,and the immunoregulatory effects of its enzymatic products were studied.The peony seed protein was extracted by alkaline extraction and acid precipitation method and the extraction conditions were optimized.Mice were used as experimental animals,and different doses of peony seed proteolysis were used for gavage.The immune regulation of peony seed proteolysis products on mice was evaluated through the determination of multiple immune indicators in mice.At the same time,the peony seed proteolysis product was used as an adjuvant to mix with the ptfA gene vaccine of Pasteurella multocida avian vaccine.Detect the level of humoral and cellular immune response induced by it and the protective effect of challenge,determining the effect of peony seed proteolysis products on the immune effect of Pasteurella ptfA gene DNA vaccine.The results showed that,the optimal technology combination of peony seed protein extraction is extraction time of 100 min,extraction temperature of 55 ?,p H11,and material-liquid ratio of 1:20(m:V).Under this process combination condition,the protein extraction rate reached 85.89%;After gavage at different doses,the immune parameters of the mice in the gavage group were higher than those in the control group.Among them,the immune organ index,DTH response,peritoneal macrophage phagocytosis ability,serum hemolysin level,spleen lymphocyte proliferation level and IFN??,IL-2 and IL-4 secretion levels of mice in the middle and high dose groups were significant Very significantly higher than the control group.When used peony seed proteolysis product as adjuvant,it had no significant effect on serum antibody levels induced by the ptfA gene DNA vaccine of Pasteurella multocida,but it can increase the proliferation of peripheral blood lymphocytes and the secretion of IL-2 and IL-4 in immunized chickens.And it provided higher protection rate for experimental animals than ptfA gene DNA vaccine without adjuvant.It was shown that the peony seed proteolysis product had a good immune regulation effect on mice,and used it as an adjuvant could enhance the immune effect of ptfA gene DNA vaccine to a certain extent.
