In this study,alfalfa polysaccharide was used as the soft template to prepare nano selenium by biological reduction of sodium selenite(biological method)by Bacillus sp.110-2.The effects of optimized culture conditions on the formation of nano selenium were discussed.On this basis,the antioxidant activity of selenium polysaccharide in vivo and in vitro was discussed.1.The extraction rate of Alfalfa Polysaccharide was 18.9%±0.24%,and the molecular weight was 9783Da.The composition of Alfalfa Polysaccharide was relatively simple,mainly including arabinose,xylose and galactose,glucose and rhamnose,with the molar ratio of9:11.3:9.8:1:1.2.Bacillus sp.110-2 has a good tolerance to Na2Se O3,and nano selenium can be reduced in the medium containing Na2Se O3.3.Through response surface methodology,the optimal culture components for synthesizing nano selenium Alfalfa Polysaccharide were 5%sucrose,1%yeast powder and 0.1%urea.The optimal culture conditions were as follows:the initial p H was 7 and the rotation speed was160 R/min at 40?for 36 h.4.Alfalfa Polysaccharide has good antioxidant capacity,but the antioxidant capacity of selenium Alfalfa Polysaccharide is higher than that of Alfalfa Polysaccharide.5.Selenidated Alfalfa Polysaccharide can effectively improve the symptoms of alcoholic liver injury in mice;it can control the liver body ratio of mice,and has a certain protective effect on the liver of mice;it can improve the activities of SOD,GSH PX and GSH in the liver;it can reduce the content of MDA in the liver;the levels of alt,AST,TG and TC in serum are decreased;the contents of TNF-?and IL-1?in liver tissue are also increased Lower.
