The purpose of this study was to explore the biological functions of 4-methylguaiacol.In this paper,we studied the antioxidative effect of 4-methylguaiacol on AAPH-induced oxidative stress in LO2 and its effect on the autophagy activity.We also evaluated the biological function of 4-methylguaiacol by determining its indicators on alcoholic liver injury and anti-oxidation.To provide relevant reference for the research on autophagy and medicine of polyphenol compounds,the main conclusions were as follows:(1)The quantitative analysis of 4-methylguaiacol in liquor samples by GC-MS was conducted to study the content range of 4-methylguaiacol in liquor.LO2 were pretreated with different concentrations of4-methylguaiacol,then AAPH induced oxidative stress in LO2.The concentrations of ROS,MDA,GSH,SOD,CAT and GSH-Px in the cells damaged by oxidation were measured.The experimental results showed that 4-methylguaiacol could significantly reduce the content of intracellular ROS and MDA,increase the activity of antioxidant enzyme.Its antioxidant capacity was gradually increased with the increase of the concentration of 4-methylguaiacol.4-methylguaiacol can reduce the oxidative damage of LO2 at the concentration range of 0?4 mmol/L.(2)The LO2 cells were treated with 4-methylguaiacol at different concentrations.Western Blot,RT-PCR and cellular immunofluorescence techniques were used to observe the effect of 4-methylguaiacol on the autophagy activity.The experimental results showed that after LO2 cells were treated with4-methylguaiacol at different concentrations,compared with the control group,the LC3 protein expression and the mRNA expression of LC3 protein was gradually increased while the p62 protein expression was gradually decreased with the increase of 4-methylguaiacol concentration.The confocal microscope observation showed that 4-methylguaiacol could significantly induce the occurrence,development and maturation of autophagy under both nutrition and starvation conditions.4-methylguaiacol can up-regulate the autophagy activity of LO2 at the concentration range of 0?4 mmol/L.(3)The mice were intragastrically administered with 50% edible alcohol for consecutive eight weeks to establish an alcoholic liver injury model,and at the same time,they were intragastrically administered with different doses of 4-methylguaiacol to investigate its protective effects on alcoholic liver injury in mice.The results showed that compared with the model group,the AST,ALT,TC,and TG of the mice given4-methylguaiacol by gavage were significantly improved.The activities of SOD,CAT,GSH-Px and GSH in the liver tissue homogenates of the mice after gavage with 4-methylguaiacol were higher than those in the model group.4-methylguaiacol had protective effects on alcoholic liver injury in mice and the effects were dose-dependent.
