Turmeric?Curcuma longa L.?,is a traditional perennial herb that can be used as medicine and food.It is widely cultivated in the southeast and southwest regions of China.Because of its effects of relieving menstruation,expelling cold and inflammation,dispersing wind and activating blood circulation,it is mostly used in the field of traditional Chinese medicine.As the main active ingredient of turmeric,curcumins have the functions of anti-oxidation,bacteriostasis,anti-inflammatory,anti-cancer and lipid-lowering and liver protection effects.As a natural pigment,it has been shown good antioxidant and whitening effects in the cosmetics industry.Due to the important role of curcumins in the health field,the market demand for curcumins is increasing.In recent years,there has been more and more research on the application of curcumins in health foods.This paper mainly discussed the anti-oxidation,anti-cancer activity and curative effect of curcums from curcuma from Hebei Province.The main research contents and results are as follows:Based on the above tests and their results,this study summarizes the following conclusions:?1?A cell damage model of H2O2 injury HepG2 was established,and the change of cell survival rate under different doses was measured by MTT.The results showed that the cell survival rate of the model group was only?75.3±3.5?%,with the increase of the drug concentration,and the cell survival rate increases.When the concentration was 20 mg/L and 40 mg/L,the cell survival rate has almost no effect compared with the blank group.The change of cell morphology can be seen:after the establishment of the H2O2 injury HepG2 cell model,compared with the blank group,the cell morphology of the model group was obviously destroyed,and the cells were concentrated,rounded,apoptosis occurred.After the drug administration,the morphology of cells was improved,and the cell morphology remained good when the concentration was 40 mg/L.Through the measurement of cellular ROS content,it was concluded that curcumins can reduce the increase of intracellular ROS content after injury,exert the characteristics of anti-oxidative stress;Through the determination of SOD,CAT and GSH-Px,it was shown that curcumins can effectively slow down the oxidative damage caused by H2O2 stimulated cells,and shoed a dose-dependent relationship with the drug concentration.The inhibitory effect of the three enzyme activities was most obvious when the concentration was 40mg/L.The determination of intracellular antioxidant provides a theoretical basis for subsequent in vivo experiments.?2?According to the measurement of the inhibition rate of cell activity,curcumins had a significant inhibitory effect on HepG2 cells in a dose-dependent and time-dependent manner,and its IC50 value was 129.02 mg/L for 24 h;It was found that curcumin-like compounds can destroy the cell structure and play a role in promoting apoptosis after HepG2 cells.According to the cell migration inhibition experiment,the migration inhibition effect is proportional to the time and dose.When the administration effect was72 h at a concentration of 150 mg/L,the inhibition effect of cell scratch distance reaches90%.With the increasing concentration of curcumin added,Hoechst 33258 stained HepG2cells were stained with high fluorescence intensity;The number of early and late apoptotic cells in HepG2 cells treated with different dose-dependent concentration increaseded.When the concentration was 50 mg/L and 100 mg/L,the proportion of viable cells was59.8%and 35.3%,the early apoptosis rate was 40.1%and 59.1%,and the apoptosis rate was significant?p<0.05?.When the concentration was increased to 150 mg/L,the early and late apoptosis rates were 79.8%and 7.9%,and the effect of promoting apoptosis was better than that of the positive control group.?3?Through the measurement of various indexes of alcohol liver injury model mice,it was found that the liver index of the model group mice increased by 18%compared with the blank control group,which indicated that the liver swelling was caused by alcohol gavage,the liver mass and index all increased;The results of the serum and liver homogenate of the mice in each group,showed that the activity of ALT in the serum of the low,medium,and high dose groups was significantly reduced compared with the model group,indicating that curcumins had a certain effect on liver function in mice with liver injury;The concentrations of TC,TG and LDL-C in serum were significantly reduced,and the concentration of HDL-C in the high-dose group was significantly increased,indicating that curcumins can improve fat metabolism function;GSH-Px,SOD,CAT and liver were significantly increased,but MDA content in liver tissue was reduced.They all showed that curcumins can improve the ability of liver antioxidant system,and reduce cell membrane lipid peroxide MDA content.?4?Through the establishment of a model of chronic liver damage in mice caused by CCL4,the results showed:after gavage of CCL4 liver injury,the living physiological state had an adverse reaction,loss of appetite,dry and dull hair,and the body weight growth rate was slow down;The important indicators of liver disease showed that curcumins can play a role in relieving chronic liver injury in mice.The content of ALT and AST in the model group was significantly higher than blank group,indicated that CCL4caused severe liver damage,and the liver damage model was successfully prepared.After the administration of the drug,their contents were decreased;At the same time,the results of various biochemical indicators in the serum of the mice showed that the activities of SOD,CAT,GSH-Px and otherantioxidant enzymes in the model group all reduced,and the activity of the antioxidants enzymes is significantly increased performanced the best effect after the high-dose administration;In addition,compared with the model group,the serum lipid contents of TC,TG,HDL-C and LDL-C in the middle and high groups were significantly different and improved;The results of biochemical indexes of liver homogenate can also be seen:compared with the model group,every index showed a certain improvement trend.Compared with the blank group,CAT and GSH-Px contents showed no significant difference of the high-dose group.It indicated that curcumins had a better effect on improving liver tissue of the experimental group.For SOD vitality,there was no significant effect compared with the model group,and the significant difference was only showed when the dose was increased to a high concentration?200 mg/kg?.
