Current hangover agent research is built on soluting wine poison,dredging courage,clearing the hot and humid,eliminating phlegm,and lining stagnant.hangover agent can achieve the purpose of hangover by strengthen liver detoxifyand enzyme action and diuresis function,or prevent ethanol absorbtion of digestive tract,reduce damage to liver of ethanol and promote liver cell regeneration,ease poisoning symptoms.Aim of this study is research effect of corn peptide and isoflavone on ADH activity that can provide theoretical basis on Hangover field..1.Hydrolysis condition of corn protein powder by the single factor experiment and response surface test is optimize.The optimal hydrolysis condition is as follows:hydrolysis temperature is56℃, hydrolysis pH is9.1,hydrolysis time is2h. Under the optimal Hydrolysis condition, the degree of hydrolysis is21.82%.2.Extract condition of Flos puerariae isoflavone by the single factor experiment and response surface test is optimize.The optimal extract condition is as follows:extract temperature is86℃, Alcohol density is75%, extract time is2h,ratio of solid to liquid is1:21(g/mL). Under the optimal extract condition,extraction ratio is9.750mg/g.3.Extract condition of puerarin isoflavones by single factor and orthogonal test is optimize.The optimal extract condition is as follows:microwave is6min, microwave power is400W, ratio of solid to liquid is1:20(g/mL), ultrasonic time is40min. Under the optimal extract condition,extraction ratio is11.123mg/g.4.The experiment of influence of corn peptide,Flos puerariae isoflavone and puerarin isoflavones on alcohol dehydrogenase activity is determined through method of Vallee&Hoch.The results show that influence of three substances on ADH is promote. But activation rate of puerarin isoflavones on ADH was significantly lower than corn peptide and Flos puerariae isoflavone. So we study corn peptide and Flos puerariae isoflavone compound.The experiment of activation rate on ADH showed that when the ratio of corn peptides and Flos puerariae isoflavone is1:0.25, the activation rate of Corn peptides-Flos puerariae isoflavone compound is the maximum value(74.45%).It is close to activation rate of Corn peptides(76.52%), and27.36%higher than the activation rate of Flos puerariae isoflavone. This experiment showed that synergistic effect of Corn peptides-Flos puerariae isoflavone on ADH in vitro is not obvious.The experiment of Corn peptide,Corn peptide-Flos puerariae isoflavone,Flos puerariae isoflavone in mice is determined too.The results show that these three substances have significant anti-drunk and sober function.The reason is that these three substances can improve the activity of ADH. The experiment of influence on ADH on Kunming mice through method of Vallee&Hoch show that activation rate on ADH of corn peptide ishighest, then is Corn peptides-Flos puerariae isoflavone compound. This experiment showed that synergistic effect of Corn peptides-Flos puerariae isoflavone compound on ADH in mice is not obvious, but this compound have significant anti-drunk effect.5.The experiment of removing DPPH radical showed that the ratio of corn peptides and Flos puerariae isoflavone is1:0.25, the clearance rate of Corn peptides-Flos puerariae isoflavone compound is the maximum value(77.83%).It is close to Corn peptides(78.43), and14.21%higher than clearance rate of Flos puerariae isoflavone(63.62%). On this ratio clearance rate of DPPH radical is high, activation rate on ADH is high too.
