The mixed juice,as one of the favorite beverages of residents all over the world,is favored by consumers because of its rich nutrients and certain health benefits.In this study,ginger,grapes and carrots were selected as the main raw materials,and a single factor experiment and response surface experiment were optimized to develop a functional mixed juice.The in vitro antioxidant capacity,edible safety evaluation and intervention were carried out.The main research results of the toxic effects of arsenic are as follows:1.Preparation of the mixed juice of ginger,grape and carrot and determination of its in vitro antioxidant capacity.Through single factor experiment and response surface experiment optimization design,the sensory score of the mixed juice of ginger,grape and carrot is used as the evaluation index,and the sensory scoring standard of the mixed juice is used to study the composition ratio of the mixed juice of ginger,grape and carrot.According to the sensory quality of juice,the optimal ratio of the mixed juice of ginger,grape and carrot was finally determined as follows:ginger juice addition amount 8.90%,grape juice addition amount 23.20%,carrot juice addition amount 14.70%,xylitol addition amount 5.00%,citric acid addition amount 0.10%,the rest is water,add appropriate amount of xanthan gum.After testing the antioxidant capacity of the mixed juice of ginger,grape and carrot in vitro,it is found that the mixed juice has strong in vitro antioxidant capacity,and its scavenging rate of hydroxyl radicals(·OH)and ABTS free radicals(ABTS+·)the removal rate is proportional to the concentration of the mixed juice.2.Safety evaluation of the mixed juice of ginger,grape and carrot.According to the"Technical Specification for Health Food Inspection and Evaluation",a 30-day feeding experiment of mouse with the mixed juice was carried out.The results showed that after 30 days of continuous intake of16.65 m L/kg·BW the mixed juice of ginger,grape and carrot,the mouse's average body weight,average daily drinking water,organ weight,organ coefficient and serum biochemical indicators had no significant changes compared with the control group,indicating that the mixed juice has no obvious toxicity to mouse.In order to better evaluate the edible safety of the mixed juice,on this basis,this study conducted a 60-day feeding experiment in mouse.The results showed that after continuous intake of the mixed juice for 60 days,the mouse's average body weight,average daily drinking water,organ weight,organ coefficient,serum biochemical indexes of liver function,oxidative stress indexes of liver and kidney tissues,and histopathological observations compared with the control group,there is no significant change,which shows that the mixed juice of ginger,grape and carrot will not cause significant effects on mouse and is a safe and non-toxic product.3.Intervention effect of the mixed juice of ginger,grape and carrot on arsenic-induced toxicity in mouse.ICR male mouse were free to drink sodium arsenite aqueous solution containing 10 mg/L arsenic,and the experimental exposure period was 60 days.At the same time,the mixed juice at the dose of 16.65 m L/kg·BW was given by gavage,once every other day,to explore intervention effect of the mixed juice of ginger,grape and carrot on arsenic-induced toxicity in mouse.The experimental results show that the mixed juice of ginger,grape and carrot has a certain alleviating effect on the toxic effects of arsenic in mouse,which is reflected in the significant activity of serum aspartate aminotransferase(AST)and alanine aminotransferase(ALT)in mouse after the mixed juice intervention it can also significantly reduce the inhibition of arsenic on the total superoxide dismutase(T-SOD)activity of mouse liver,kidney and testis tissues and significantly increase the content of glutathione(GSH)in mouse kidney and small intestine tissues.The induced increase of malondialdehyde(MDA)content in liver and testis tissues and hydrogen peroxide(H2O2)content in liver tissues has a significant inhibitory effect,and it also reduces the structure of the above-mentioned main organs and tissues of mouse with arsenic intake from drinking water damage.The above results show that the mixed juice of ginger,grape and carrot prepared in this study helps to alleviate the toxic effects of arsenic in mouse,and has a certain protective effect on the main organs and tissues of mouse.
