Arsenic(As)is a non-metallic element widely found in nature.The main sources of environmental arsenic pollution included natural factors and human activities.Excessive levels of arsenic in the environments can adversely affect human health and the environment system.Arsenic enters the human body mainly through food and drinking water.Arsenic entering the body can cause damage to various tissues and organs,and induce diseases such as nervous system,cardiovascular system,respiratory system,digestive system and reproductive system.Arsenic exposure was linked with human skin cancer,lung cancer and liver cancer,and so on.Previous studies have shown that the use of a certain amount of vitamin C,vitamin E or selenium compounds can antagonize the toxic effects of arsenic on experimental animals,but so far,there have been few reports on the use of dietary regulation to interfere with arsenic toxicity.Therefore,we tried to interfere with the arsenic toxicity in experimental animals by using the cereal breakfast powder and grape pigment.The following research results were obtained:(1)Establishment of the animal model of chronic arsenic poisoning via drinking water in mice.Tested mice were given the drinking water containing10 mg/L of arsenic for 60 days.The results showed that arsenic intake caused significantly decreases in superoxide dismutase activity and glutathione content,and increases in the levels of hydrogen peroxide and malondialdehyde in mouse kidney,lung,testis and heart tissues,accompanied with the tissue lesions in the kidney,lung and testicular tissue.The quality and the quantity of sperm were decreased in As-exposed group mice.(2)Intervention of breakfast powder on As toxicity was carried out.The model mouse of arsenic poisoning via drinking water established above was used.The animals feeded with drinking water containing 10 mg/L arsenic,were given intragastrically 7.15%(g/m L)or 14.30%(g/m L)of breakfast powder for 60 d.The results showed that the breakfast powder intake can reduce the contents of hydrogen peroxide and malondialdehyde in kidney,lung,testis and heart tissue,increase of superoxide dismutase the activity and glutathione content in the tissues,and have some protective effects against As-caused tissue structure damage.Compared with the arsenic exposure group,the sperm quality and the total sperm count were increased after intragastric administration of breakfast powder.The results showed that the intervention of multigrain breakfast powder can alleviate the oxidative damage and toxic effects of arsenic.(3)Intervention of grape skin pigment to arsenic toxicity was tested.The established animal model of arsenic poisoning via drinking water was used.The tested mice with drinking water containing 10 mg/L arsenic for 60 days,were given 180 mg/L grape skin pigment at the same time.It was found that grape pigment can alleviate oxidative damage of arsenic to kidney,lung,testis and heart,reduce the content of hydrogen peroxide and malondialdehyde in above tissues,and increase the superoxide dismutase activity and glutathione content.And also the total number of sperms was increased,the rate of sperm deformity was decreased.The grape skin pigment showed a protective effect against As-caused tissue structure damage.(4)Intervention of different concentrations of grape skin pigment to arsenic toxicity was studied.Using the established mouse model of arsenic poisoning via drinking water containing 10 mg/L arsenic for 56 days,the tested mice were allowed 2.5 mg/ kg·bw or 4.5 mg/kg·bw grape pigment by intragastric administration.The results showed that both concentrations of grape pigment can alleviate the oxidative damage of arsenic to mouse kidney,lung,testis and heart.The grape pigment at 4.5 mg/ kg·bw significantly decreased the contents of hydrogen peroxide and malondialdehyde in kidney,lung,testis and heart tissue,increase superoxide dismutase activity and glutathione content,reduce As-caused increases in the contents of total cholesterol,triglyceride content,and in the activities of aspartate aminotransferase and alanine aminotransferase.It can also increase the sperm count,reduce sperm deformity rate,and reduce the damage to tissue structure caused by arsenic.
