Yellow rice wine is a traditional Chinese wine with great historical and cultural value,it was once known as the "national wine".With the increase of people's demand for health,yellow rice wine develops towards the direction of low alcohol content and high nutrition,and enjoys the name of "liquid cake" because of its health promoting value.Further confirmation of the nutrition and health value of yellow rice wine requires more scientific evidence,so the focus of research has been shifted from food processing,physical and chemical properties to its health function.Up to now,few research focused on the intervention effect of rice wine on aging model.In this study,the d-galactose-induced aging mouse model was established to analyze the related effect of rice wine on aging body and explore its mechanism.These would provide a preliminary research basis for further exploring of the beneficial effect of traditional Chinese yellow rice wine.The main results are as follows:1.Effects of yellow rice wine on the cognitive ability of D-gal induced aging mice.The D-gal induced aging mouse model were established and the effects of different doses of yellow rice wine on their cognitive ability were evaluated by measuring body weight,hair,antioxidant indexes,general behavioral state,cholinergic system and apoptotic gene expression in hippocampal tissue of mice from each experimental group.The results showed that:(1)The six week subcutaneous injection of D-gal into the cervical ridge of mice resulted in hair removal,dull and coarse hair,slow movement,slow reaction and less weight gain;the activity of antioxidant enzymes,the brain and organ indexes of the model mice were significantly decreased,while the content of lipid oxidation products significantly increased,suggesting that the aging model was successfully established.(2)The study found that the influence of yellow rice wine intake on aging mice was dosage-dependent.low dose of yellow rice wine did not improve the general aging symptoms and antioxidant capability of aged mice significantly,while moderate and high doses of yellow rice wine relieved the general aging symptoms of D-gal induced aging mice and improved the brain and organ index of aging mice.(3)Both low,moderate,and high doses of yellow rice wine can improve the antioxidant capacity of the aged mice,and among them the moderate dose of yellow rice wine exhibits the best effect.(4)Moderate yellow rice wine intake can reduce the cognitive impairment of Dgal induced aged mice to a certain extent,which may be related to the improvement of the cholinergic system in the hippocampus and the inhibition of nerve apoptosis.2.The effects of yellow rice wine on anti-fatigue ability of D-gal induced aging mice.The anti-fatigue effects of moderate yellow rice wine intake on D-gal induced aging mice were evaluated by measuring the resting time of mice during the forced swimming experiment and the content of glycogen in liver and serum content of lactic acid and urea nitrogen after exercise.The results showed that:(1)D-gal induced aging mice were more prone to fatigue during exercise compared with normal mice.(2)Different doses of the rice wine intake showed different effects on the anti-fatigue ability of aging mice.While medium and high dose of yellow rice wine can significantly improve the anti-fatigue ability of aged mice by increasing their liver glycogen storage and removing metabolites effectively during exercise.3.Effects of yellow rice wine on intestinal flora of D-galactose-induced aging mice.Metagenomic analysis of intestinal flora in mice faeces was performed by high-throughput sequencing,the results showed that:(1)D-galactose-induced aging mice showed significant changes in intestinal microflora structure,compared with normal mice.For the diversity of microflora was significantly increased,and the proportion of Firmicutes/Bacteroidetes was significantly changed.After the intervention of moderate amount of yellow rice wine,the diversity and F/B values of the aged mice showed a more similar pattern to those of the normal control group,suggesting that the intestinal microbial community of the aged mice could be improved to some extent.(2)The moderate intake of yellow rice wine increased the relative abundance of butyric acid producing bacteria in the intestinal microflora of aging mice,which could help maintain the balance and stability of the intestinal environment and delay the aging process.In conclusion,moderate intake of yellow rice wine could improve the cognitive ability,the anti-fatigue ability of aging mice during exercise and their microecology of intestinal flora.This study preliminarily explored the molecular mechanism underlying the effects of traditional Chinese rice wine on aging.Especially,revealed the beneficial effects exhibited by moderate intake of yellow rice wine.These results could be helpful to improve our understanding of the drinking value of Chinese traditional yellow rice wine and to promote the development of yellow rice wine towards a more healthy and nutritious direction.
