Study On The Anti-aging Activity Of Indigo Chinensis And Preparation Of Oral Liquid

Lonicera edulis L.is an all-natural wild edible berry.Lonicera edulis L.is rich in anthocyanins,polyphenols and other bioactive substances and has anti-tumor,hypolipidemic and other health care functions.However,there are few studies on the anti-aging function of Lonicera edulis L..With the improvement of national consumption level,the basic material demand has been fully guaranteed,healthy life has become a new standard to measure the happiness of life,and the social cognition of health has gone beyond "no disease is healthy",delaying aging is of great significance to meet the current national pursuit of health.In this project,Lonicera edulis L.was used as experimental material and D.melanogaster as model organism to verify the effect of Lonicera edulis on delaying aging,explore the mechanism of Lonicera edulis L.on delaying aging and develop Lonicera edulis functional food.Firstly,the anti-aging effect of Lonicera edulis pulp on male and female D.melanogaster was determined by exploring the effects of Lonicera edulis pulp on their life span,climbing ability,food intake and reproductive ability.The results showed that the average life span of female D.melanogaster was extended by 12.64% and11.08% in low and medium dose groups(2 and 5 mg/m L),respectively.The results showed that a certain dose of Lonicera edulis pulp could prolong the life of female D.melanogaster,while the same dose of Lonicera edulis pulp had no significant effect on the life of male D.melanogaster.It can significantly improve the climbing ability of male and female D.melanogaster,reduce the reproductive ability of D.melanogaster,improve the stress resistance of male and female D.melanogaster,improve the antioxidant activity of male and female D.melanogaster,reduce the level of lipid metabolites in male and female D.melanogaster,and reduce the weight of male and female D.melanogaster.It can be concluded that Lonicera edulis pulp can delay the aging of female D.melanogaster and improve the physiological function of male D.melanogaster to a certain extent.After successfully verifying the anti-aging effect of Lonicera edulis pulp,the anti-aging mechanism of Lonicera edulis pulp was explored by extracting total m RNA of D.melanogaster and PCR.The results showed that d Fox O gene and m TORC1 gene in the dose group were significantly decreased compared with the blank control group,suggesting that Lonicera edulis pulp may delay D.melanogaster senescence through insulin and m TOR pathways.Finally,through sensory evaluation and other methods,the functional oral liquid of Lonicera edulis pulp was developed.The optimal formula was determined as 80% of Lonicera edulis pulp,12% of sugar,0.6% of ascorbic acid and 0.6% of carrageenan.The best sterilization conditions were determined as 90?,20 min.The content of polyphenols and anthocyanins in the oral liquid of Lonicera edulis pulp were determined to be 1.551±0.006 mg/m L and 2.293 ±0.076 mg/m L.In conclusion,Lonicera edulis pulp can delay D.melanogaster senescence.The experimental results of this subject can not only provide an important theoretical basis for further elucidating the mechanism of the nutritional components of Lonicera edulis pulp in anti-aging,but also provide an important basis for the development of new safe anti-aging food and drugs.
