China has abundant flaxseed acid-containing plant resources,and Xinjiang is also the hometown of safflower.In recent years,many researches have been done on the nutritional composition,content,and functional effects of linseed oil and safflower seed oil.Previous research found that flaxseed and safflower seed blend oil(FSSBO)made according to a certain proportion of scientific compatibility has a good blood lipid lowering function.Therefore,based on the previous research on FSSBO for lowering blood lipids,this article further conducts research on the effect and mechanism of FSSBO on blood glucose in animal models of type ? diabetes.First,the intervention effect of FSSBO on blood glucose in mice was preliminarily discussed.Second,to explore different ratio flaxseed,safflower seed oil affect ? diabetes mice blood sugar;Third,to explore FSSBO affect ? diabetes mice blood sugar;Fourth,to explore the hypoglycemic mechanism of FSSBO.The main research contents and conclusions are as follows:(1)First of all,using flaxseed oil as a raw material,on the basis of the preparation of flaxseed oil extract by the integration of saponification and purification in the early stage of the research group,a new process for the preparation of flaxseed oil extract by high-speed shear saponification-freezing separation method was developed,Streptozotocin(STZ)was used to establish a mouse diabetes model,and the successfully modeled mice were randomly divided into hyperglycemia control group,safflower seed oil group,linseed oil group,and linseed oil extract group,FSSBO group.The test mice were orally administered with the test substance in groups for 14 days.The blood glucose-lowering effect of FSSBO was preliminarily analyzed from mouse body weight,glucose tolerance,and insulin level Indicators.(2)Based on the preparative preparation of safflower hypolipidemic soft capsules by the research group,it was found that the ratio of 60%linseed oil extract and 40%safflower seed oil has the best comprehensive lipid-lowering effect.High-fat and high-sugar milk gavage combined with small intraperitoneal multiple injections of STZ method to establish a mouse model of type 2 diabetes,linseed oil and safflower seed oil were given at different ratios and doses to verify the effect of lowering blood sugar at this ratio.The results showed that 60%linseed oil extract Compounding with 40%safflower seed oil can improve the islet ?-cell secretion function and insulin resistance in diabetic mice.(3)Based on the combination of 60%linseed oil extract and 40%safflower seed oil(FSSBO),which has a good blood glucose lowering effect,again explore the synergy of the effects of polyunsaturated fatty acids on blood glucose in type 2 diabetic mice.Comparing the hypoglycemic effect of FSSBO with safflower seed oil,linseed oil,and linseed oil extract,the results showed that FSSBO had the best hypoglycemic effect,increased insulin secretion,and increased HOMA-?(P<0.05),Improve HOMA-IR(P<0.01)(compared with the hyperglycemia model group,HOMA-IR is reduced by 60.71%),improve the body's sensitivity to insulin,and thereby improve the symptoms of glucose and lipid metabolism disorders in type 2 diabetic mice,reaching a reduction Blood sugar effect.(4)Western Blot was used to detect the expression of p-PI3K,p-GSK-3? and p-Akt protein in the liver of rats,so as to preliminarily grasp the mechanism of FSSBO's hypoglycemic effect.Compared with the model group,the weight of FSSBO group was generally increased,but there was no statistical significance(P>0.05),fasting blood glucose and liver weight index were significantly reduced(P<0.05 or P<0.01),p-GSK-3? protein expression was effectively down regulated(P<0.05),p-PI3K and p-Akt protein expression was up regulated,but there was no statistical significance(P>0.05).
