In order to further understand the material basis of healthy effects of Allium mongolicum,based on high performance liquid chromatography-mass spectrometry(HPLC-MS),combined with animal metabolism experiments,the key characteristic metabolites of Mongolian chive were deeply explored,and the contents of key metabolites in water extracts of Allium mongolicum from different habitats were detected and compared.In the first part,rats were given 2 ml of water extract of 3:100 Mongolian chive by gavage.Blood samples were collected at 0.5 h,1 h,2 h and 4 h after administration,and LC-MS was used Detection methods: the chemical components of rat serum samples and water extract of Mongolian chive were analyzed respectively.Through the comparative analysis of serum components of rats before and after administration,and the comparison of chemical components between serum samples and Mongolian chive,the differential metabolites directly from Mongolian chive in bleeding plasma were screened.The secondary spectra of these compounds were further obtained by LC-MS / MS,and one of them was identified as alliin by database matching and standard comparison.In the second part,based on HPLC detection technology,the absolute content of alliin in Allium mongolicum from three different producing areas and planting methods was determined.The results showed that there were significant differences in alliin content in several samples from different sources.The alliin content of Mongolian chive planted in Inner Mongolia was the highest(29.8 mg / g),while that of Gansu was only 15.8 mg / g.All in all,alliin was found to be one of the most important components in Allium mongolicum,and it was stable in the process of metabolism.On this basis,the determination of alliin content revealed that there might be differences in nutritional components of alliin from different producing areas and planting methods.At the same time,an HPLC method for the determination of alliin content in Allium mongolicum was established.
