Soybean is an important grain and oil crops,rich in high quality protein,oil and other bioactive substances.Soybean polyphenols,being secondary metabolites during soybean growth,can scavenge free radicals as a kind of natural antioxidant foods.It also has hypoglycemic,hypolipidemic,bacteriostatic,anti-inflammatory and anti-tumor effects.Therefore,Soybean polyphenols have long captured the interests from researchers in the fields of nutrition,medicine and food science.In this study,cultivated soybean(Glycine Max)and wild soybean(Glycine soja)were used as materials to explore the differences in the content of polyphenol active components and antioxidant activities between wild and cultivated soybeans.At the same time,the optimal conditions for the purification of soybean polyphenols were explored and liposomes were prepared.The main results are as follows:1.By analyzing the contents of polyphenols,flavonoids and isoflavones of wild soybean before and after germination and their antioxidant capacity in vitro,it was found that the contents of polyphenols,flavonoids and isoflavones and antioxidant capacity of wild soybean after germination were significantly increased compared with that before germination.2.In this study,the contents of polyphenols,flavonoids and isoflavones in 16 kinds of cultivated soybeans and 3 kinds of wild soybeans were analyzed.Meanwhile,DPPH,ABTS,FRAP methods and ROS scavenging ability were used to explore the difference in antioxidant capacity of 19 kinds of soybean sprouts.The results showed that the content of polyphenols and antioxidant capacity of wild soybean sprouts were higher than that of cultivated soybean.Wild soybean GDW20 had the highest total polyphenol content and antioxidant activity,flavonoids and phenolic acids are accounted for 98.01% of the total polyphenols content,and were the main components of wild soybean GDW20 sprout polyphenols.3.The optimal purification conditions of AB-8 macroporous resin for wild soybean GDW20 soybean sprout polyphenols were explored.The results showed that the adsorption and the elution performance of the macroporous resin was the best when the sample concentration was 3 mg/mL,the pH was 3 and the concentration of ethanol eluent was 75%.4.The inclusion of polyphenols by polyphenol liposomes can significantly reduce cytotoxicity,increase the survival rate of oxidative damaged cells,decrease intracellular reactive oxygen species and decrease mitochondrial membrane potential.Moreover,the results of cell uptake test showed that the cells could effectively absorb polyphenol liposomes which had good lysosomal escape function when entering the cell and played a role in the cytoplasm.In conclusion,this study indicates that wild soybean has high polyphenol content and antioxidant activity as an excellent resource for soybean polyphenol extraction.Liposome can improve the efficiency of soybean polyphenols,providing a reference for the future application of soybean polyphenols.
