With the rapid development of society and the increase of people's work pressure,most people are in a sub-health state.Insomnia,obesity,aging,diabetes and lipid metabolism disorders often occur.People hope to improve this phenomenon through daily diet.Therefore,at the present stage,we need to develop a product with improved sleep and antioxidant properties to meet the needs of the vast number of people.In this paper,semen ziziphi spinosae was combined with Lanzhou lily,Ophiopogon japonicus,gardenia and spring Amomum villosum to develop a functional beverage.Its technological formula and storage quality were studied,and its mechanism of improving sleep and antioxidant activity were discussed.The research contents and conclusions of this paper are as follows:(1)Study on the preparation of semen ziziphi spinosae compound stock solution and its flavor substances by ultrasonic-assisted water decoctionTaking the extraction rate of total saponins and total flavonoids as the index,the response surface optimization test was carried out on the basis of single factor test results.The optimum process conditions were as follows: ultrasonic power 250 W,ultrasonic time 26 min,decocting temperature 90 ?,decocting time 50 min and decocting times twice.The content of total saponins and total flavonoids in the stock solution prepared by ultrasonic-assisted water decoction is higher than that prepared by water decoction.The flavor substances in the stock solution prepared by the two processes were compared by gas phase ion mobility spectrometry(gc-ims).It was concluded that the types of flavor substances in the stock solution prepared by the two processes were less different,and the concentration of most flavor substances in the stock solution prepared by ultrasonic-assisted decoction was higher.(2)Optimization of technological formula and antioxidant activity of compound stock solution of semen ziziphi spinosaeThrough orthogonal test combined with fuzzy mathematical sensory evaluation method,the formula of compound stock solution of semen ziziphi spinosae was optimized.The best formula was obtained as follows: 2.0g of semen ziziphi spinosae,2.0g of Lanzhou lily,1.5g of Ophiopogon japonicus,1.5g of Gardenia and 1.0g of spring Amomum villosum.Taking the centrifugal sedimentation rate as the index,the homogenization process of stock solution is studied.The optimum homogenization conditions are as follows: homogenization pressure 30 MPa,homogenization time4 min and homogenization times twice.Taking the total number of colonies and sensory score as indicators,the sterilization process of stock solution was studied.The best sterilization conditions were as follows: sterilization with pressure cooker at 121 ?for 15 minutes.The antioxidant activity,physical and chemical indexes,sensory indexes and microbial indexes of semen ziziphi spinosae compound stock solution were determined.The results showed that it had strong antioxidant activity,and all indexes met the national standards.Using the zero order kinetic model and Arrhenius equation,the shelf-life prediction model of sensory score value is established.It is predicted that the shelf-life of stock solution is about 194 days at 25 ?.(3)Network pharmacological study on the mechanism of semen ziziphi spinosae compound in improving sleepThrough the method of network pharmacology,the mechanism of semen ziziphi spinosae compound improving sleep was studied.The results showed that the active components of semen ziziphi spinosae compound improving sleep mainly included saponin A,spinozin and Ophiopogon japonicus flavone,which mainly acted on targets such as ESR1,HTR2 C and TNF Regulation of calcium signaling pathway and estrogen signaling pathway to improve sleep.(4)Study on Optimization of technological formula and antioxidation of semen ziziphi spinosae compound beverageThrough the single factor test and taking the sensory score as the index,the best formula of the beverage is as follows: 20% thyme juice and 0.6% compound sweetener(stevioside: rock sugar is 3:1).Taking the centrifugal precipitation rate and viscosity as the indexes,through single factor and orthogonal optimization test,the optimum addition amount of thickener is as follows: Xanthan Gum 0.06%,galactomannan 0.04%,CMC Na0.06%.Taking the centrifugal sedimentation rate as the index,the best homogenization conditions are obtained through single factor test:homogenization pressure 30 MPa,homogenization time 3min and homogenization times twice.Taking the total number of colonies and sensory score as indicators,the best sterilization conditions were obtained through single factor test: sterilization with pressure cooker at 121?
