In this paper,the fluorescence properties of apomorphine,O-nornuciferine and magnoflorine belonging to apomorphine alkaloids were studied.A new fluorescent method was established for the determination of apomorphine in Apomorphine Hydrochloride Sublingual Tablet.And new fluorescent methods were established for the determination of magnoflorine in Chinese herbal medicines Clematidis Radix et Rhizoma(Weilingxian)and Ziziphi Spinosae Semen(Suanzaoren).Three aspects are included in this thesis:1.The endogenous fluorescence and sensitized fluorescence of apomorphine were studied.It was found that the addition of borax,SDS in apomorphine solution or pure methanol solvent can significantly enhance the fluorescence of apomorphine.Based on these three fluorescence enhancement methods,new fluorescence methods for the determination of apomorphine in the Apomorphine Hydrochloride Sublingual Tablet were established,and the results of the new methods were 3.29,3.18 and 3.35(mg/tablet),respectively.The results were verified by an ultraviolet spectrophotometry and found to be 3.25(mg/tablet),consistent with the results of the fluorescence method.2.The fluorescence properties of O-nornuciferine were studied.It was found that the fluorescence of O-nornuciferine was weak in aqueous solutions,but it was enhanced when the aqueous solution with higher methanol volume fraction.The fluorescence quantum yields of near-neutral and alkaline aqueous solutions with 70% methanol were measured to be Y = 0.10 and 0.049,respectively.The study found that the SDS solution has a better sensitizing effect on the fluorescence of O-nornuciferine,which can increase Y to 0.19.3.Weilingxian and Suanzaoren are two common Chinese herbal medicines,and magnoflorine is one of the active ingredients of the two herbs.From the three-dimensional fluorescence spectra of the extracts of Weilingxian and Suanzaoren,it was found that the coexisting components of the two herbs had no effect on the fluorescence peak of magnoflorine.Based on this,the content of magnoflorine in the two herbs were determined by fluorometric analysis,and the results were 0.131% and 0.161%.The three-dimensional fluorescence combined with second-order calibration method was also used to analysis the sample and the results obtained were 0.130% and 0.158%.Finally,the above two methods were verified by HPLC-MS method,and the results were 0.128% and 0.160%,which proved that the fluorescence method proposed in this thesis were reliable.
