| After"Healthy China"became a national strategy,China’s big health industry entered a period of rapid development.Plant protein with its natural,green,health,nutrition and other characteristics,has won the favor of more and more consumers.In order to integrate the industrial resources of Perilla Perilla and improve its strategic position in the health industry,this paper took Perilla cake meal as raw material and extracted the protein of Perilla meal(PSMP)through the combination of alkali extraction and foam separation method.The basic properties of alkaline extraction process,foam separation process and PSMP were studied.The results were as follows:(1)PSMP was extracted from Perilla Perilla seed meal by alkali extraction.Combined with single factor and response surface optimization experiments,the optimal extraction conditions were as follows:p H9,liquid-solid ratio of 500:1 m L/g,extraction time of 8 min,extraction temperature of 35℃.Under these conditions,the extraction rate of PSMP was 48.5%.(2)Glycine betaine(GB)was added in the PSMP foam fractionation process.The results of UV,FTIR,fluorescence and scanning electron microscopy showed that the two compounds interacted with each other through cation-π.Increasing the concentration of GB leads to the decrease of the surface tension of PSMP-GB solution,and the enhancement of the foaming property and foam stability of the solution,which is conducive to the adsorption of PSMP on the bubble surface.The recovery R and enrichment ratio E of 0.3 g/L GB were increased by 26%and 1.4 times,respectively,compared with that without GB.When must ensure that GB concentration change of solution p H,in the process of p H rise,PSMP-GB solution increases,the surface tension of the solution of the foaming ability and foam stability is abate,strengthened the drainage process,p H is equal to the 7 R was 83.6%,the recovery of adsorption densityΓis 1.96 mg/m~2.It is indicated that GB can be used as a kind of green and natural foam reinforcing agent.(3)In general foam fractionation device to add a component within the regular-hexagon hollow prismoid(RHP),by comparing with a RHP component,enrichment and recovery of the two RHP R than E and the change of the air bubble diameter,it is concluded that join components will improve the PSMP-GB enrichment than E,the enrichment ratio E along with the increase of gas velocity,liquid volume is reduced,in order to get a high concentration of concentrate,but by reducing gas velocity,decrease in liquid to achieve purpose.In order to obtain proper recovery R and high enrichment ratio E,the amount of RHP can be appropriately reduced.In conclusion,the addition of RHP in the fractionation tower can effectively improve the foam phase discharge process and the fractionation efficiency of the foam.(4)Sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)diagram showed that foam fractionation PSMP showed four clearer bands;PSMP was composed of 17kinds of amino acids including 7 kinds of essential amino acids by HPLC,which indicated that foam fractionation could effectively separate PSMP.PSMP has good functional properties,and has a good market prospect in the field of food processing and cosmetics research and development. |
