As an important part of Chinese civilization,traditional Chinese medicine(TCM)has made remarkable contributions to the prosperity of the Chinese nation and exerted a positive influence on the progress of world civilization.Chinese patent medicine(CPM),which has unique advantages in carrying,storing,transporting,taking and so on,is a kind of TCM preparation made from Chinese herbal medicine according to prescription and technology.With the advantages of strong specificity,good stability and good reproducibility,TCM fingerprint has been widely used in quality evaluation of CPM.Fluorescence spectroscopy has the characteristic of high sensitivity,and three-dimensional fluorescence fingerprint can better reflect the overall characteristics of CPM.Three-dimensional fluorescence fingerprints of 11 kinds of CPMs were studied in this paper.The characteristic parameters of three-dimensional fluorescence spectrum data were extracted using Matlab software and the consistency of three-dimensional fluorescence spectra of CPMs was investigated using cluster analysis and principal component analysis.The three-dimensional fluorescence fingerprints of CPMs with good consistency of fluorescence spectra were established by the average method.The fluorescence components corresponding to the fluorescence peaks in three-dimensional fluorescence fingerprints were identified by comparing with the fluorescence spectra of standards.The paper consists of five parts.1.Introduction.The status of the research of CPMs with three-dimensional fluorescence spectroscopy,the cluster analysis and principal component analysis combined with three-dimensional fluorescence spectroscopy and 11 kinds of CPMs were reviewed with 264references in this part.2.Investigation of experimental conditions of fluorescence spectroscopy.The fluorescence spectra of CPMs were measured by fluorescence spectroscopy.The conditions of fluorescence spectrometer and sample extraction were investigated and optimized,and the reliability of the method was verified by methodological experiment.The optimal instrument conditions are as follows,response time 0.5 s,negative high pressure 700 V,scan speed 2400nm·min-1,slit width 5.0/5.0 nm.The sample extraction conditions are as follows,ultrasound temperature 25℃,ultrasound time 15 min for Qingre Sanjie capsules and Jinlianhua capsules and 10 min for the other 9 kinds of CPMs,and methanol content 100%.The methodological experiment shows that the proposed method has good repeatability and stability.3.Study on three-dimensional fluorescence fingerprints of four kinds of CPMs for Shugan Lidan.Under optimum experimental conditions,three-dimensional fluorescence spectra of Xiaoyan Lidan tablets,Hugan tablets,Lidan Paishi tablets and Jindan tablets were determined.The consistency of fluorescence spectra was investigated by chemometrics method,respectively.The results show that the differences of three-dimensional fluorescence were observed between Guangdong Baihe and the other three manufacturers.The differences were also observed for three-dimensional fluorescence spectra of Jindan tablets produced by two manufacturers.The three-dimensional fluorescence spectra of Hugan tablets and Lidan Paishi tablets are all in good consistency,respectively.The three-dimensional fluorescence fingerprints of CPMs with good consistency of fluorescence spectra were established by the average method,respectively.The preliminary results were obtained by comparing with the fluorescence spectra of the standards:Xiaoyan Lidan tablets contain caffeic acid and chlorogenic acid.Hugan tablets contain schisandrin,schisandrin A and schisandrin B.Jindan tablets contain resveratrol and polydatin.The fluorescence peaks of Lidan Paishi tablets are not matched with the fluorescence peaks of the standards and the fluorescence components of Lidan Paishi tablets can not be confirmed.4.Study on three-dimensional fluorescence fingerprints of four kinds of CPMs for Qingre Jiedu.Under optimum experimental conditions,three-dimensional fluorescence spectra of Jinlianhua capsules,Qinghuo tablets,Qingre Sanjie capsules and Fufang Chuanxinlian tablets were determined.The consistency of fluorescence spectra was investigated by chemometrics method,respectively.The results show that the three-dimensional fluorescence spectra of 4 kinds of CPMs are all in good consistency,respectively.The three-dimensional fluorescence fingerprints of 4 kinds of CPMs were established by the average method,respectively.The preliminary results were obtained by comparing with the fluorescence spectra of the standards:the fluorescence peaks of 4 kinds of CPMs are not matched with the fluorescence peaks of the standards and the fluorescence components of CPMs can not be confirmed.5.Study on three-dimensional fluorescence fingerprints of three kinds of CPMs for Zhike Pingchuan.Under the optimum experimental conditions,the three-dimensional fluorescence spectra of Fufang Jiegeng Zhike tablets,Keteling capsules and Qingfei Yihuo tablets were determined.The consistency of fluorescence spectra was investigated by chemometrics method,respectively.The results show that the good consistency of three-dimensional fluorescence spectra of Fufang Jiegeng Zhike tablets produced by five manufacturers was observed.The three-dimensional fluorescence spectra of Qingfei Yihuo tablets produced by Yunnan Tengyao and Xiuzheng are all in good consistency.The three-dimensional fluorescence spectra of Qingfei Yihuo tablets produced by Yunnan Jinke and Yunnan Qujing have good consistency,respectively,but there are some differences from Yunnan Tengyao and Xiuzheng.The three-dimensional fluorescence spectra of Keteling capsules produced by three manufacturers are different.The three-dimensional fluorescence fingerprints of CPMs with good consistency of fluorescence spectra were established by the average method,respectively.The preliminary results were obtained by comparing with the fluorescence spectra of the standards:Fufang Jiegeng Zhike tablets contain caffeic acid and chlorogenic acid.The fluorescence peaks of Keteling capsules and Qingfei Yihuo tablets are not matched with the fluorescence peaks of the standards,and the fluorescence components of Keteling capsules can not be confirmed.
