Fingerprint refers to the chromatogram or spectrogram that can indicate the chemical characteristics of some complex substances by using certain analytical methods.Fingerprint is a method for quality inspection that shows the characteristics of fingerprints as a whole.It was achieved that the international consensus for the quality analysis of traditional Chinese medicines?TCMs?and its preparations was fingerprints.There are broad application prospects in the research and quality evaluation of Chinese patent medicines?CPMs?by fingerprints.The fluorescence properties and fluorescence spectra were investigated by fluorescence spectroscopy in this dissertation for four types of CPMs including Liqi Zhitong,Yangxue Yiqi,Xiaozhong Zhitong and Runchang Tongbian medcines,and the experimental conditions were optimized.The consistency was investigated with cluster analysis and principal component analysis for the three-dimensional fluorescence spectrum of CPMs from different manufacturers and different batches of the same manufacture,and the all data and the extracted characteristic parameter data of the three-dimensional fluorescence spectra were used.The three-dimensional fluorescence fingerprints were established by the average method for each kind of CPMs and the experimental data can be used for the quality evaluation of CPMs.This dissertation consists of six chapters.Chapter one:Introduction.The research status of the application of three-dimensional fluorescence spectroscopy combined with cluster analysis or principal component analysis,and the research status of 14 kinds of CPMs were reviewed.The purpose and significance of this dissertation were described briefly.The 151 references were cited.Chapter two:Investigation of experimental methods of fluorescence spectrum.The experimental conditions for fluorescence spectrometry were optimized with the Huoxue Zhitong capsule as sample and the optimum experimental conditions were obtained.The scanning speed,slit,response time and negative high voltage were selected as 2400nm·min-1,5.0/5.0nm,0.5s and 700V,respectively.The precision,repeatability,stability of the fluorescence spectrometry and the fluorescence properties of the samples were investigated,and the results proved that the proposed method was accurate and reliable.Chapter three:Study on Three-dimensional Fluorescence Fingerprints of CPMs for Liqi Zhitong.The experimental conditions were optimized for the determination of fluorescence spectra of CPMs for Liqi Zhitong.Cluster analysis and principal component analysis were applied to the investigation of the similarity and difference of three-dimensional fluorescence spectra of CPMs for Liqi Zhitong.The comparison results showed that the clustering and principal component analysis results with extracted characteristic parameter data can better reflect the consistency and difference of three-dimensional fluorescence spectra.The three-dimensional fluorescence spectra of Yinxingye tablets,Guanxin Danshen capsules and Fufang Danshen tablets from different manufacturers were in good agreement.The three-dimensional fluorescence spectra of Baoxinning capsules from the two manufacturers were different.According to the results of Cluster analysis and principal component analysis,the three-dimensional fluorescence fingerprints of CPMs for Liqi Zhitong with good consistency were established by the average method,respectively.Chapter four:Study on Three-dimensional Fluorescence Fingerprints of CPMs for Yangxue Yiqi.The experimental conditions were optimized for the determination of fluorescence spectra of CPMs for Yangxue Yiqi.Cluster analysis and principal component analysis were applied to the investigation of the similarity and difference of three-dimensional fluorescence spectra of CPMs for Yangxue Yiqi.The comparison results showed that the three-dimensional fluorescence spectra of CPMs for Yangxue Yiqi were well consistent.According to the results of Cluster analysis and principal component analysis,three-dimensional fluorescence fingerprint of CPMs for Yangxue Yiqi with good consistency were established by the average method,respectively.Chapter five:Study on Three-dimensional Fluorescence Fingerprints of CPMs for Xiaozhong Zhitong.The experimental conditions were optimized and selected for the determination of fluorescence spectra of CPMs for Xiaozhong Zhitong.Cluster analysis and principal component analysis were applied to the investigation of the similarity and difference of three-dimensional fluorescence spectra of CPMs for Xiaozhong Zhitong.The comparison results showed that the clustering and principal component analysis results with extracted characteristic parameter data can better reflect the consistency and difference of three-dimensional fluorescence spectra.The three-dimensional fluorescence spectra of Fufang Sanqi capsules and Sanqi tablets from different manufacturers were in good agreement.The three-dimensional fluorescence spectra of Huoxue Zhitong capsules of Jiangxi Baishen and other manufacturers were different.According to the results of Cluster analysis and principal component analysis,three-dimensional fluorescence fingerprint of CPMs for Xiaozhong Zhitong with good consistency were established by the average method,respectively.Chapter six:Study on Three-dimensional Fluorescence Fingerprints of CPMs for Runchang Tongbian.The experimental conditions were optimized and selected for the determination of fluorescence spectra of CPMs for Runchang Tongbian.Cluster analysis and principal component analysis were applied to the investigation of the similarity and difference of three-dimensional fluorescence spectra of CPMs for Runchang Tongbian.The comparison results showed that the clustering and principal component analysis results with extracted characteristic parameter data can better reflect the consistency and difference of three-dimensional fluorescence spectra.The three-dimensional fluorescence spectra of Runchang capsules and Dahuang Tongbian capsules from different manufacturers were well consistent.The three-dimensional fluorescence spectra of Tongbianling capsules from Wuhan Kangle and other manufacturers were different.According to the results of Cluster analysis and principal component analysis,three-dimensional fluorescence fingerprint of CPMs for Runchang Tongbian with good consistency were established by the average method,respectively.
