Chiral compounds,chiral medicine,significance and methods of optical resolution,high performance liquid chromatography,p-hydroxyphenylglycine,and enantioselective membranes were introduced.Four chiral stationary phases(CSP)of telcoplanin were obtained.(1)The CSP-1was prepared by bonded telcolanin to surface of silica gel with isocyanate propyl triethoxysilane linker;(2)The synthesis of CSP-2 was that teicoplanin,methacrylate isocyanate and sulfhydryl silica gel were reacted successively;(3)The CSP-3 was to introduce the end-enyl group into teicoplanin,and then to react with mercapto silica gel;(4)Preparation of CSP-4 was glyceryl ether oxygen trimethoxysilane as the bonding arm.Those stationary phases were used for chiral separations,which possess enantioseparation efficiency,especially for α-amino acids and their derivatives.Among those columns,more enantiomers were separated on CSP-1,however,CSP-2have the best optical resolution to some compounds.The solubility of two enantiomers of p-hydroxyphenylglycine was determined by high performance liquid chromatography.The effects of temperature and p H on their solubility were studied.From the solubility curve,it is confirmed that the p-hydroxyphenylglycine is a racemic compound.The chiral composite membrane of teicolanin was prepared with1,6-diisocyanate and teicolanin by interfacial polymerization,and the membrane extraction performance of teicolanin against p-hydroxyphenylglycine was studied.We tested the extraction effects of solvent,amount of chiral selector,extraction time,sample concentration and temperature.The orthogonal experiment was designed to determine the optimal experimental conditions.When methanol was used as the eluent,the amount of teicolanin was 2%,the extraction time was 40 min,the sample concentration was 0.02 mg/m L,and the extraction temperature was 10℃,the chiral membrane extraction could reach 63.8% enantiomer excess value.
