| Seed,working as an extremely important germplasm resource and capital goods in the agricultural sector,plays a vital guiding role in the development of agriculture.As an important oil and economic crop in China,peanut has low reproductive coefficient(about 1:10)and large amount of seeds used in production.Therefore,high-vigorous seeds were one of the important factors to ensure high seedling rate,plant uniformity and production potential in peanut.The aging of seeds after harvest not only affects the storage of quality,but also causes the seed vigor,the germination index and nutrient quality decreasing gradually,and finally results in the yield loss of peanut production and processing.It is of extraordinary significance to explore the seeds vitality of peanut for the full utilization in industry and agriculture.Here,six lines,including Hua414,Hua 606,Hua 825,Hua 9102,Hua8107,ZP06,were selected to evaluate their seed vigors using natural aging,conductivity method,TTC content method and artificial aging method.Many traits related to seed vigor,such as seed germination rate and germination index,were carried out as well as SDS-PAGE electrophoresis.The main findings are as follows:1.In the different stages of natural aging of peanut seeds at 3,6,9,12,15,18,21 months,the oleic acid content of Hua 414,Hua606,Hua825,Hua 9102,Hua8107 and ZP06 decreased first.The trend of increasing and then decreasing;the linoleic acid content showed the opposite trend,and the wave shape changed at 6 months;the seed germination rate,germination potential,germination index,growth amount,vigor index decreased significantly;the conductivity increased significantly High,TTC content decreased significantly,among which Hua414,Hua 825,Hua9102 and Hua8107 were more vigor and more resistant to storage than Hua606 and ZP06.2.The artificial aging methanol chemical method was used to detect the seed vigor of different peanut varieties.The results showed that the seed vigor of each variety gradually decreased with the prolongation of methanol treatment time.When aging for 15 min,the germination index decreased sharply,with a drop of 30%~60%,Hua 414 and Hua 9102 have the lowest decrease,Hua 606 germination rate has the largest decrease;when methanol aging for 55 min,Hua 414 and ZP06 can not germinate;in high temperature and high humidity environment,seed germination rate and other indicators with aging treatment time Increasing and decreasing,the germination rate of each variety decreased by 15%~22.5% when aging for 1 day;when aging for 3 days,the germination index of each variety decreased sharply,and all varieties could not germinate when aged for 5 days.3.The protein extraction method and protein expression profile of different vigor peanut varieties in the germination stage showed that the extraction of seed protein by petroleum ether method is a better protein extraction method;through the sister line Hua 8107(normal germination rate)and ZP06 SDS-PAGE electrophoresis analysis(low germination rate)showed that the protein content of the two varieties was significantly different at 23 k D,25 k D,26 k D,45 k D,and the content of 8107 was higher than that of ZP06.With the prolongation of germination time,the protein content of the two varieties gradually increased at 23 k D,25 k D,26 k D,45 k D.reduce. |
