EST-SSR Development And Genetic Diversity Research On Zanthoxylum Based On RNA-seq

Zanthoxylum bungeanum(pricklyash),which is a type of condiment and cultived broadly is a valuable medicinal and economic tree in China.Nowadays,ample of pricklyash which their origin are vague causing lots of problems.On the basis of RNA-seq,both Z.bungeanum and Z.armatum were sequenced in this research.By right of their transcriptome data,EST sequencing and SSR locus are analysing as key points,which including screening of EST-SSR primers,genetic diversities and ties of consanguinity.As a result,this study try to explain some questions about interspecific relationship and intraspecific relationship of Zanthoxylum whose contribution provide crucial datum support for identification and inheritance.1.After sequenced by high throughout platform,as is shown: Producing more than0.03 billion clean reads per sample and 0.258 billion reads totally.Specifically,more than 4G clean bases data were detected,meanwhile,there were 38.76 G clean bases in total and the accuration of sequencing reached 95%(Q20)and 90%(Q30).Additionally,more than0.245 billion reads and 36.76 G clean data generated in all.2.There are 64,944 Unigene sequences in Zanthxoylum bungeanum where there are12,746 SSR locus distributing in 10,595 Unigene whose frequency of occurrence is16.31%,and a SSR loci appearing every 4.24 kb.For Z.armatum,15,096 SSR locus distribute 12,613 Unigene which there are total 75,669 Unigene.The frequency of occurrence is 16.67% and a SSR loci appears every 3.91 kb.3.There are 26 pairs EST-SSR primers manifest excellent polymorphism which have been detected in Zanthoxylum.4.6 pairs EST-SSR primers are selected to identify ties of consanguinity and genetic diversities of 125 samples Zanthoxylum.The result shows Z.bungeanum and Z.armatum can be identified accurately in interspecific crossing while can not distinguish in intra-species.5.There are manifest genetic differentiation between Z.bungeanum and Z.armatum.On the contray,There are little genetic differentiation and gene diversities in intra-species especieslly in varieties and also don’t have significant regionalism.
