| In this study,the typical representative variety of the national geographical indication product Huaiyuan white flower jade pomegranate(Punica granatum L.)was selected as the initial research material.The study was conducted through different clone propagation methods,different culture conditions,and different methods of squeezing seedlings,transplanting and cutting seedlings of tissue culture seedlings.Taking the research path of tissue culture seedling transplant survival rate calculation,growth index,root scanning,physiological and biochemical index detection and cost accounting,etc.,to further explore the impact of different cultivation conditions on the establishment of white flower jade pomegranate group cultivation seedling system;and seedlings Quality and seedling cost accounting.The aim is to provide a theoretical basis for the selection of white flower jade pomegranate seedling propagation methods and factory breeding seedlings,so as to improve the reproductive capacity and application promotion level of high-quality pomegranate seedlings in Huaiyuan.The test results are as follows:1.In this experiment,the stem tip was used as the test material to obtain the white flower jade pomegranate tissue culture seedlings.The best condition for sterilization of the stem tip as an explant was 0.1%HgCl2 for 10 min;the best primary culture medium was WPM+NAA 0.75 mg/L+6-BA 0.5 mg/L+35 g sucrose+5.5 g agar;the best subculture medium is WPM+NAA 1.0 mg/L+6-BA 1.0 mg/L+35 g sucrose+5.5 g agar;The optimal rooting induction medium is WPM+IBA 0.8 mg/L+35 g sucrose+5.5 g agar.2.It is concluded from the research on the conditions of smelting seedlings that with a certain range,the survival rate of smelting seedlings continues to increase with the increase of light intensity and humidity,and a downward trend will appear when the critical value is reached,so the optimal light intensity is 1000-1200 LX,the optimal relative humidity is 65-75%;it is concluded from the research on the conditions of refining seedlings that the selection of appropriate fungicide types and substrate formulations can significantly improve the survival rate of transplanted tissue culture seedlings,up to 85.56%.3.This study compared the growth,root system,physiological and biochemical indicators of the transplanted annual white flower jade pomegranate tissue culture seedlings with the same period of annual cutting seedlings.The results showed that the tissue culture seedlings were superior in root vitality and growth and development activity For cutting seedlings,referring to the actual production,it is calculated that the production cost per plant is less than 5 yuan/plant,and the cost will gradually decrease with the increase in the number of tissue culture seedlings.The cost can be reduced to 1.69 yuan/Strain,when the scale production reaches 100,000 plants.In summary,the tissue culture nursery approach of pomegranate can be used as a way to produce seedlings on a large scale.According to the market sales price of cutting seedlings(20±2 yuan/plant),the profit of two-year-old tissue culture pomegranate seedlings can reach more than 8 yuan,so the group cultivation method can be used as a factory breeding method,which has a broad Promotion prospects. |
