In this study,wild Anoectochilus roxburghii was collected from ningde,nanping and sanming areas of fujian province to isolate and identify its mycorrhizal.The R11(Fusarium concentricum)strain was selected to establish a symbiotic system with the Anoectochilus roxburghii,Observing and measuring the growth of Anoectochilus roxburghii in the greenhouse.Using high-throughput sequencing technology to analysis the Anoectochilus roxburghii that infected R11(Fusarium concentricum),to explore the effect of mycorrhizal fungi on the growth of Anemone roxburghii by the change of gene expression.The experimental results are as follows:(1)Endophytes of the Anoectochilus roxburghii in ningde,nanping and sanming areas of fujian province which belong to16 genera and include 9 fungi and 10 bacteria were isolated from the roots of healthy wild plants.(2)Fusarium concentricum on the Growth of plantlets was analysed through six-month-culture in the glasshouse.The Anoectochilus roxburghii inoculated with R11(Fusarium concentricum)increased in root length,survival rate,number of leaves and fresh weight compared with uninoculated Anoectochilus roxburghii,eecially in root length,number of leaves and The fresh weight increased significantly;inoculation of R11(Fusarium concentricum)mycorrhizal fungi can effectively increase the net photosynthetic rate of Anoectochilus roxburghii.(3)The transcriptome was sequenced from the inoculated leaves of Anoectochilus roxburghii,and 76,418 unigenes were obtained by sequence licing,of which 37278unigenes were annotated.Comparing the number of differential genes before and after inoculation,there were 508 differentially expressed unigenes in the 28d inoculation,of which 174 were up-regulated and 334 were down-regulated.There were 40 differentially expressed genes between the inoculation and the control.According to the gene GO analysis,the results of differential showed gene enrichment in pigmentation and nutrient reservoir activity was the most obviously,followed by growth,antioxidant activity,cell junction and extracellular region part.According to the enrichment analysis of KEGG pathway,the most significant pathways were photosynthesis-antenna proteins,flavonoid biosynthesis,circadian rhythm-plant,phenylpropanoid biosynthesis and glycolysis.The above results will provide theoretical basis for plant-bacteria interaction mechanism,and provide scientific reference for the application of mycorrhizal fungi in Anemone roxburghii production practice.
