Anthocyanins are important flavonoids that belong to phenylpropanes and rep resent a large class of plant secondary metabolites,which involve many aspects of normal plant growth and development,and help plants resist biotic and abi otic stresses.Studies have shown that environmental factors such as light,horm ones and sucrose affect the accumulation of anthocyanins in plants,thus impro ving plant stress resistance.At present,there are few studies on anthocyanin sy nthesis in Camellia oleifera.This experiment studied the effects of environment al factors on anthocyanin accumulation in secondary cotyledons of Camellia ole ifera,and the effects of stress induced by anthocyanins on the growth and dev elopment of secondary cotyledons of Camellia oleifera.It can provide a theoret ical basis for stress-resistant breeding of anthocyanins in Camellia oleifera.the embryogenic calli(called secondary cotyledons)induced from Camellia ol eifera seeds were used as experimental materials to explore the effects of envir onmental factors such as light,hormone and sucrose on anthocyanin accumulati on in secondary cotyledons of Camellia oleifera.On this basis,the growth inde x,chlorophyll content,soluble protein,soluble sugar,malondialdehyde,superoxi de anion,proline and other physiological and biochemical indexes of Camellia oleifera secondary cotyledons under different treatments were compared.The eff ects of stress on the growth and development of secondary cotyledons of Cam ellia oleifera were further analyzed.The results are as follows:1.Increasing light intensity and prolonging photoperiod promoted anthocyanin accumulation and anthocyanin synthase expression in secondary cotyledons of Camellia oleifera.2.Increasing sucrose concentration had no effect on anthocyanin content in s econdary cotyledons of Camellia oleifera,but significantly increased the express ion of anthocyanin synthase.3.Abscisic acid and methyl jasmonate promoted anthocyanin accumulation a nd anthocyanin synthase expression in secondary cotyledons of Camellia oleifer a,which reached the peak on the 12 th day,and then began to decrease.4.Adding methyl jasmonate,sucrose,increasing light intensity and prolongin g photoperiod could promote the growth and development of secondary cotyled ons of Camellia oleifera,but after too long time,it began to inhibit its growth and development.The content of osmotic regulation substances(soluble sugar,soluble protein,etc.)and the activities of antioxidant enzymes(SOD,POD,et c.)increased at first and then decreased.5.The addition of abscisic acid seriously affected the growth and developme nt of Camellia oleifera secondary cotyledons,the growth index decreased,and t he chlorophyll content was almost zero.The content of osmotic regulation subs tances and the activities of antioxidant enzymes were significantly increased.
