| Panax ginseng C.A.Meyer is one of the precious traditional Chinese herbal medicines and has high medicinal value.Due to its dormant properties,it brings many troubles to the production of seedlings.Therefore,in this experiment,3-4 years old ginseng were used as the experiment material.According to the characteristics of ginseng’s own dormancy,the method to breaking the dormancy of ginseng and the changes in the content of medicinal ingredients in different parts of ginseng during the growth process has been explored.The results showed:1、Different hormone types,concentrations and treatment time had different effects on breaking Ginseng dormancy.In this experiment,50 mg.L-1GA3solution was used to soak the roots for 10 h to broke the dormancy of ginseng.The germination rate of ginseng was 79.53%,the germination days were 7 days,the leaf development days were 23 d,and the plant height was 12.92 cm.2、The combination of different low temperature treatments and hormone treatments had the best effect on breaking ginseng dormancy.After 30 d of low temperature treatment at-2~-3℃,combined with 50 mg.L-1GA3solution for 10 h,the germination rate of ginseng was the highest,89.96%,the germination days were 4d,the leaf development days were 17 d,and the plant height was 13.56 cm.3、Ginseng was stored in different environments during the dormant period.After being taken out,it was treated with low temperature and then combined with hormone treatment.The results of the study found that the growth index of Ginseng stored in matrix environment was higher than those of ginseng stored without matrix.It works best when stored in matrix environment during dormancy.4、Ginseng sprouts had different changes in the medicinal components in the body under different growth days.As the growth time of Ginseng sprouts progresses,the content of total saponins and flavonoids both reached the maximum at 55 days of growth.Then the content of total saponins decreased,and the content of flavonoids tended to stabilize;The medicinal components of Ginseng sprouts above-ground were of higher value,and the medicinal components of Ginseng sprouts stored with matrix were higher than those without matrix.5、During the growth process of Ginseng sprouts,the changes of superoxide dismutase(SOD)activity and malondialdehyde(MDA)content are the same.Both show a maximum value at 55 days of growth,and then decrease.The SOD activity and MDA content of the underground part Are higher than the above-ground part;The changes of peroxidase(POD)and catalase(CAT)activities are the same.They both have a maximum value at 25 days of growth,and the activity of the above-ground part is higher than that of the underground part.The difference is POD activity showed an upward trend at 35-65 d,while CAT activity continued to decrease after the maximum value.The antioxidant enzyme activity and MDA content of Ginseng sprouts preserved with substrate were higher than those without substrate. |
