Pre-harvest sprouting(PHS)of wheat was a worldwide disaster that seriously influence the yield and quality of wheat.PHS was influenced by many factors,such as dormancy,grain color etc.Many QTLs associated with PHS have been mapped on wheat all 21 chromosomes.QTLs on group 3 and 4 homologous chromosomes have been identified frequently.This study was based on the QPHS.sicau.3D chromosome identification of PHS resistance sites of SHW-L1(synthetic wheat).On this basis,this study successfully developed 14 pairs of STS markers at this locus(570.4-572.0Mb,referring to China Spring(CS)genome v1.0),and analyzed the relationship between the genetic differences and germination rate of 250 common wheat accessions.Through the phenotype identification of the germination rate of 250 common wheat accessions seeds,the average germination rate was 73.32% in 7 days.High-resistance PHS(GR:0-10%)accounted for 6.80% of the total materials,and the average germination rate was 3.64%.Resistance PHS(GR: 10-30%)accounted for 6.40% of the total number of materials,and the average germination rate was 20.11%.Middle-resistance PHS(GR: 30-60%)accounted for 10.00% of the total materials,the average germination rate was 45.29%.Sensitive PHS(GR: 60-100%)accounted for 76.80% of the total materials,the average germination rate was 87.57%.According to the identification results of 250 common wheat accessions,it is divided into 3 haplotypes,of which the average germination rate of Haplotype Ⅰ was40.88%,and 48 accessions were collected in total.The average germination rate of Haplotype Ⅱ was 73.48%,and 17 accessions were collected in total.The average germination rate of Haplotype Ⅲ was 81.72%,and 185 accessions were collected in total.Using the 14 pairs of STS markers developed by the PHS resistance locus,24 accessions PHS resistance materials with PHS resistance locus were screened out.These markers will help to use this resistance locus to screen PHS resistance materials.The resistance materials with resistance locus screened out at the same time can provide a reference for breeding workers.
