Sanghuang is a precious medicinal fungus.Its main active ingredients are polysaccharides and brassic substances.It has good adjuvant therapeutic effects in anti-tumor.It is currently recognized as the best anti-cancer drug by scholars at home and abroad.Fungus is used,but there are problems such as slow growth of Sanghuang fruit bodies and low medicinal effect content,especially in terms of protoplast regeneration,which is seriously lower than other edible and medicinal fungi.In this paper,Sanghuang mycelium is used as the material,and the protoplasts are first prepared.Then carry out natural regeneration and corona electric field to mulberry mutagenesis and regeneration respectively,and screen good regenerating colonies for independent cultivation,in order to finally obtain a Sanghuang mutagenic strain with a faster growth rate and higher medicinal effect than the original strain,and use the regenerated strain Carry out artificial cultivation to cultivate Phellinus linteus fruiting bodies.The experimental results are as follows:1.The cell wall of the mycelium of Sanghuang was digestified with2% lysozyme at 25 ℃--30 ℃ for 3 hours to achieve a better demystification effect.There is no obvious linear relationship between the yield of protoplasts and the mycelium age.2.The maximum regeneration rate of protoplasts was 0.05%-0.065%,and the growth rate of mycelium of the protoplast regenerating strains was variable.After sequencing,it was found that the variation occurred inside the base.3.When the electric field treatment time is 40 s,it has obvious promotion effect on the growth rate of Sanghuang regenerative hyphae,and the average daily growth is 1% higher than that of the control group;when the electric field treatment time is 30 s,the electric field has an effect on the growth of Sanghuang regenerative hyphae It has a strong inhibitory effect,and the maximum daily growth rate is 16% lower than that of the control group.4.The protease activity of the regenerated strain of strain zsh increased after 10 minutes of electric field treatment,and the protease activity of the regenerated strain of strain zhs-1 increased after 40s electric field treatment,which increased by 2.8% and 6.8%,respectively,and the growth rate of the strain It is consistent with the level of protein activity,and it can be seen that the protease activity is positively correlated with the growth rate of mycelium;The activity of extracellular amylase of strain zhs-1 treated with corona electric field for 30 seconds increased by 57% compared with the control group,while the activity of the other group of mycelial amylase was weak and accompanied by slow growth of mycelium.Sanghuang mycelium growth rate is related to amylase activity.5.The growth rate of the mycelium of the regenerated strain and the original strain were obviously different under bag cultivation,and some of the strains were obtained with earlier formation of fruiting bodies.
