Stearic acid,oleic acid,linolenic acid,trans 10 cis 12 conjugated linoleic acid,cis 9 trans 11 conjugated linoleic acid and other eighteen carbon fatty acids are added to animal diets to improve lactation performance and fatty acid synthesis related genes Make an impact.The main purpose of this study is to explore the molecular mechanism of t10c12-CLA regulating milk component metabolism in goat mammary epithelial cells through sterol element binding protein 1(SREBP1).The mammary gland tissues of dairy goats were collected first,and mammary epithelial cells were isolated.Use eighteen carbon fatty acids to process breast epithelial cells,use transcriptome sequencing technology to analyze differential genes and significantly enriched signal pathways,detect protein expression levels,and combine with signal pathway inhibitors to conduct cell signal transduction Research and obtain the following results:1.Five octadecanoic fatty acids(stearic acid,oleic acid,linolenic acid,trans-10cis-12 conjugated linoleic acid,cis-9trans-11 conjugated linoleic acid Oleic acid),after 72 hours of treatment,it was found that t10c12-CLA had a significant inhibitory effect on SREBP1(P <0.05).Subsequent treatment of GMECs with 100μM t10c12-CLA and sequencing analysis revealed that there were 49 genes differentially expressed with the control group.KEGG enrichment analysis found that AMPK signaling pathway closely related to glucose and lipid metabolism and multiple genes related to lipid metabolism were significantly enriched.2.After adding 100μM t10c12-CLA to GMECs for 12 h,24h,and 72 h,it was found that the phosphorylation level of AMPK was significantly up-regulated(P <0.05)and the phosphorylation level of m TOR was significantly down-regulated(P <0.05).The expression of SREBP1 protein was significantly inhibited(P <0.05).P <0.01).After the introduction of AMPK inhibitors,it was further discovered that t10c12-CLA inhibited the expression of SREBP1 by activating AMPK.3.After adding t10c12-CLA and AMPK inhibitor Dorsomorphin to GMECs alone or in combination,it was found that the expression of β-casein in GMECs had no effect(P>0.05);lipid droplets were reduced(P <0.01),and C16 in fatty acids(P <0.01))And C18(P <0.05)content decreased,fatty acid metabolism genes were suppressed;the m RNA expression level of glucose transport related gene GLUT8 was down-regulated(P <0.05).In summary,t10c12-CLA plays a very important role in regulating the metabolism of goat milk components.It can significantly reduce the fatty acid content in goat milk and also participate in sugar metabolism.The above result may be that t10c12-CLA activates AMPK and affects SREBP1 Caused by expression.
