| Lonicerae Caulis is a dry flower bud or flower with initial bloom of Lonicera macranthoides,Lonicera glandularis,Lonicera nannanensis or Lonicera isabelli.At present,the main source of medicinal honeysuckle is Lonicera macranthoides.It is mainly distributed in Hunan,Guizhou,Chongqing,Guangxi and other places in South China,with the largest output.The branches,vines and leaves of Lonicera japonica Thunb.Were used as the prevention and treatment of infectious diseases in cattle,pigs and poultry.In this paper,by collecting the branches and vines of Lonicerae Caulis from different places of origin in China,according to the requirements of Pharmacopoeia of the people’s Republic of China(2015 Edition)on the quality of Chinese herbal medicines,we carried out the detection and analysis of properties,microscopic and powder identification,water content,extract content and chlorogenic acid content,and carried out the analysis of the antibacterial and total antioxidant capacity of the water extract and alcohol extract of Lonicerae Caulis.The following results were obtained:(1)The stem and rattan of Lonicerae Caulis from different places of origin are segmented branches and rattan,the ends of branches are often accompanied by residual leaves and leaf marks,with a diameter of 1.5-6mm;the surface is mostly smooth,brownish red,dark brown,grayish green,green or dark yellow;a few parts are hairy,the size of which is uniform,short and dense,with a length of about 1-2mm;the smooth epidermis is easy to peel off,and the outer skin is thin,with a thickness of less than 0.1mm,transparent It is brittle and easy to break,and the epidermis is not easy to break.After breaking,it will be connected because of the epidermis,and the section is yellow white and hollow;the air is tiny,with a slight fragrance of honeysuckle;the mouth is light and bitter.(2)The powder of Lonicerae Caulis is earthy yellow,brownish yellow to yellowish green;under the microscope,there are many non glandular hairs with many small bumps on the surface;the epidermal cells are yellow,yellowish green,yellowish brown to reddish brown in different shapes,irregular circle,triangle,quadrilateral or polygon,and the non glandular hairs will leave broken marks after falling off;see vacuole like stone cells;parenchyma cells.The parenchyma cells are round and have a diameter of 4-16 nm.There are a lot of calcium oxalate clusters in parenchyma cells.The results of microscopical observation on the cross-section of mature branch rattan of Lonicerae Caulis showed that: after peeling off the skin,2-4 layers of endothelial cells were irregularly arranged outside the phloem,and the endothelial cells were in the shape of quasi circular to polygonal,closely arranged;the phloem was in a circular arrangement;the cambium formed a thin layer between the phloem and the xylem,in a circular shape;the xylem was wide,in a radial arrangement,with scattered ducts;The ray passes through the whole xylem with different width and narrow;the medulla is relatively wide and composed of round to quasi round parenchyma cells,which are closely arranged.(3)According to the test method of Chinese Pharmacopoeia,the content of water is8-12% and the content of water extract is 11-16%.According to the HPLC method for the determination of chlorogenic acid in Pharmacopoeia of China,the optimized conditions were: the filler was octadecylsilane bonded silica gel,and the mobile phase was(10:90)acetonitrile: 0.4% phosphoric acid.0.28-3.02% of Lonicerae Caulis was determined with327 nm as the detection wavelength.The content of chlorogenic acid is the lowest in Chongqing,and the content of chlorogenic acid is more than 1% in other parts of Lonicerae Caulis.(4)The Oxford cup method was used to study the antibacterial activity of water extract and alcohol extract of Lonicerae Caulis to Escherichia coli and Staphylococcus aureus.The results showed that Lonicerae Caulis had obvious antibacterial effect on Escherichia coli and Staphylococcus aureus.The diameter of antibacterial ring of 1g/m L water extract to Escherichia coli was 22.07-25.01 mm,and that of 1g/m L alcohol extract to Escherichia coli was 18 Between 02-20.86 mm.The diameter of bacteriostatic circle of 1g /ml water extract to Staphylococcus aureus is between 21.06-27.61 mm,and that of 1g / ml alcohol extract to Staphylococcus aureus is between 19.05-20.95 mm.Whether it is for Escherichia coli or Staphylococcus aureus,the antibacterial effect of the aqueous extract of Lonicerae Caulis is better than that of the ethanol extract of Lonicerae Caulis.(5)FRAP method was used to determine the total antioxidant capacity of the extracts from Lonicera.The total antioxidant capacity of the extracts of Lonicera Caulis from different places is 0.807 m M-1.278 m M.Among them,the total antioxidant capacity of water extracts of Lonicera Caulis were lower than that of alcohol extracts,which was significantly higher than that of VC solution(0.6 mg / m L);the total antioxidant capacity of alcohol extract group A was the highest(1.278mm),and that of alcohol extract group B was the lowest(0.959);The total antioxidant capacity of water extraction group C was the highest(0.966mm),and that of water extraction group A was the lowest(0.807mm). |
