| The genetic background and relationship in Dianthus will provide the support for the selection of hybrid parents in future breeding and the identification of new varieties.In this study,28 plant species in Dianthus were collected from domestic and international companies.The characteristics of their seed and petals were used to analyze the genetic diversity among them.Based on the polymorphism,the SSR primers in the Dianthus transcriptome,were selected to analyze the genetic diversity at the DNA level.The results were as follows:(1)The seeds are divided into the medium-size group and the small-sizegroup in the28 plant species.All of the seeds have the raised ridges on the surface,the seed navel at the basal end of the seeds,and the striped-negative reticulate ornamentation on the seed coat surface.(2)Exception for D.Caryophyllus and D.Plumarius,D.Chinensis cv.Diamond scarlet Imp and D.Chinensis cv.Telstar.Scarlet,and Ameria and Giganteus,Cheddar Pink,D.Superbus Red,D.Chinensis,D.Bartatus Sooty,D.Chinensis cv.Red,D.Alwoodii Alpinus,D.Chinensis cv.White,D.Bartatus Red,D.Caesius,D.Badenia,D.Carthusianorum,D.Superbus,D.Deltoides Arctic Fire,D.Deltoides Confetti Cherry Red,D.Knappii,D.Cruentus,D.Monspessulanus,D.Arenarius,D.Sylvestris and D.Pavonius are completely distinguished based on the petal size,the circle at the base and the margin serrate.(3)From the 100 pairs of SSR markers obtained from the D.Chinensis transcriptome sequencing,33 pairs of primers with good generality,high polymorphism and clear amplification bands were selected.Two hundred and fifty eight bands were amplified in the 28 plant species in Dianthus.All was polymorphic and the polymorphism rate was 100.00%.Each pair of primers could amplify 4-13 polymorphic bands,with an average of 7.82 bands per pair.(4)Using NTSY-PC2.11,the genetic similarity coefficients of the 28 plant species ranged from 0.594 to 0.969.they were clustered into three major groups.The group I included all D.Chinensis except for D.Chinensis cv.Telstar.Scarlet,D.Gratianopolitanus,D.Superbus Red,D.Plumarius,D.Caryophyllus,D.Alwoodii Alpinus,two cultivars of D.Bartatus,D.Caesius,D.Badenia,D.Carthusianorum,D.Ameria,Alpine Mix(large seeds),Alpine Mix(small seeds),D.Deltoides cv.Arctic Fire.The group II included D.Knappii,D.Arenarius,D.Sylvestris,D.Monspessulanus,D.Pavonius,D.Deltoides cv.Confetti Cherry Red,D.Chinensis cv.Telstar.Scarlet and D.Cruentus.The Group III included D.Giganteus and D.Superbus.(5)The SSR molecular marker fingerprints constructed with three primers c37368_g2,c37647_g1 and c38800_g1 could distinguish all the 28 plant species in Dianthus. |
