| The inflammation is the most fundmental pathology process of infection and many diseases.Chinese herbal polysaccharides are natural,non-toxic,non residue and other special advantages;effectively avoids the toxic and side effects of chemical drugs,which has obvious development advantages for developing new anti-inflammatory and immunotoxic drugs with low toxicity and high efficiency.Glycyrrhiza polysaccharide(GP)is the main active ingredient of licorice.Recently,it has attracted more and more attention for its excellent biological activities.Selenium(Se)is an essential trace element for human beings and many other forms of life.Selenium as a kind of microelement also has the efficacy of antioxidation,immunoenhancement,participating in the formation of coenzyme Q,reducing the metal toxicity.Recently,many studies confirmed that molecular modification could greatly enhance-the biological activities or produce much new medicinal value for herbal polysaccharides,the selenzing polysaccharide(SeGUP)has important research value and application prospect.Uralensis is a common Chinese herbal medicine,it has the effect of clearing heat and removing toxicity,invigorating spleen and replenishing qi,expelling phlegm to arrest coughing with multiple.It is considered that Glycyrrhiza uralensis Fisch polysaccharide(GUP)is one of the main material bases of Glycyrrhiza uralensis Fisch.In this text,GUP was extracted from Glycyrrhiza uralensis Fisch and selenzied.The structure,the antioxidant activity,the anti-inflammatory and the immunomodulatory activity of SeGUP were investigate.Object:Molecular modification of GUP can improve its biological activities,so as to provide a theoretical basis for developing animal preparations and selenium supplement feed additives with anti-inflammatory and immune functions.Methods:1.This study was to research choosing Glycyrrhiza uralensis Fisch as the research object,using water extraction to extract the crude GUP,the removing the protein and pigment,purifying the crude Glycyrrhiza uralensis Fisch polysaccharide through the column chromatography.And GUP was obtained.The selenzing polysaccharide(SeGUP)were obtained by the modification of HNO3-Na2SeO3.The in vitro and in vivo antioxidant activity was analyzed.2.We investigated anti-inflammatory function of SeGUP in many animal models,including acute model characterized by swelling,emigration of leukocytes,capillary permeability or the chronic model characterized by hyperplasis of granulation.We also studied the effect of SeGUP took on cytokines,NF-κB and MAPKs signal transductive pathways by using RAW 264.7cells.3.The complete freund`s was used to induced arthrisis(AA)rats model.The paw swelling,arthritis index,rat weight,index of immune organs and seum inflammation factor as evaluation index,were used to evaluate the effects of SeGUP.4.By measuring the immune organ index,the carbon clearance index,the serum hemolysin level,the splenic T lymphocyte proliferation and the production of TNF-?,IL-4 and IFN-γ,we investigated the influence of SeGUP on inherent immunity and adaptive immunity in mice.5.The effect of SeGUP on chickens after immunization with ND Lasota vaccine was examed.The function of cellular immunity,humoral immunity and the activity exchange of IL-2,IL-6 and IFN-γhave been studied,in order to stduy the possible mechanisms of how SeGUP enhance immunologic function.Results:1.GUP was subsequently modified by the nitric acid-sodium selenite(HNO3-Na2SeO3)method.UV and FT-IR analysis indicated that the selenium in SeGUP existed mainly as O-Se-O and the conformation of glucose-ring kept unchanged basically.The thermal stability,particle size,molecular weight and monosaccharide composition of SeGUP differed significantly from those of GUP.Moreover,compared to GUP,SeGUP exhibited greater antioxidant activities in vitro and in vivo.2.The result showed that SeGUP(300 mg/mL)had a marked suppressive effects on normal mouse’s and ablated adrenal gland mouse’s ear edema induced by xylene,the mouse’s hind paw edema induced by carrageenin,the increased vascular permeability caused by acetic acid,the mouse’s granuloma caused by cotton ball.The result showed that SeGUP(80μg/mL)inhibited TNF-α,IL-6 and IL-1βsecretion level in a dose-dependent manner,and inhibited NO and PGE2 synthesis.The result showed that sodium ferulate inhibited NF-κB and MAPKs protein phosphorylation in a dose-dependent manner.3.SeGUP(200 mg/kg)can significantly decreased paw swelling and recovered the weight,decreased thymus and spleen index,inhibit rat peripheral blood promoting inflammatory factor levels.4.SeGUP(200 mg/kg)can enhance the immune organ index,the carbon clearance index,the serum hemolysin level,the T lymphocyte proliferation and the secretion of IFN-γ,IL-4 and TNF-αin serum.5.In vivo experiment,SeGUP could significantly promote lymphocytes proliferation at all of the time and the efficacy of SeGUP groups were better than that of GUP.The serum antibody titer of SeGUP were higher than that of vaccination control group at most of the time and were higher than that of GUP group at some times.Conclusion:To sum up,after the GUP is modified molecularly,its biological activities make an obvious change.SeGUP possessed higher bioactivities than GUP for the same dose,suggesting that selenylation modification elevated the anti-inflammatory and immunomodulatory effects of GUP. |
