| Turpan black sheep are resistant to high temperature and cold,high survival rate of lambs,and delicious meat,but the low lamb production rate and low stock volume have led to the development of reproductive performance of Turpan black sheep.Therefore,in this experiment,Turpan black sheep were selected as the test object.Fifteen non-pregnant ewes were randomly selected for the test.Morning and evening estrus tests were performed.The estrous cycle time and duration were observed and recorded to understand the estrous cycle of Turpan black sheep.On this basis,estrus identification is performed.When the estrus starts(recorded as 0 h),blood is collected every 3 h during the estrous period(0~36 h)and the post-estrous period(36~72 h).(4 d to 14 d)and pre-estrus(15 d to 17 d)blood samples were taken at 8:00 and 20:00 each day,and the supernatant was centrifuged to determine the FSH and LH in serum during the entire estrous cycle by ELISA,P4 and E2 levels,analysis of their reproductive hormone secretion rules,and provide theoretical and technical basis for further scientific research on genetics,breeding and reproductive physiology of Turpan black sheep.The test results showed that:(1)The estrus cycle of Turpan black sheep is:the average estrus interval is 15.76 d,which is mainly concentrated on 16-17 d,and the duration of estrus is 24-48 h.(2)The secretion of FSH and LH in serum throughout the estrous cycle is:the FSH concentration value(2.291 ±0.297 IU/L)during estrus is the highest,and it is significantly different from the concentration value(2.021±0.202 IU/L)during estrus.The difference(P<0.05)increased slowly during the estrus period(2.221±0.205 IU/L)until the pre-estrus period(2.240±0.237 IU/L);during the estrous period(6.945±0.488 IU/L)and the later estrus period(6.947±0.921 IU/L)is lower than the interestrus period(7.280±0.503 IU/L)and the pre-estrus period(7.472±0.559 IU/L).The concentration is generally increasing,and the pre-estrus period is significantly higher than the estrous period And estrus interval(P<0.05).(3)The secretion of P4 and E2 in serum of Turpan black sheep during the entire estrus cycle is as follows:the secretion amount is highest in the early estrus period(0.095±0.020 ng/mL),decreased during the estrous period(0.090±0.017 ng/mL),and there is no Obvious changes,secretion increased during estrus(0.094±0.014 ng/mL),the overall change was not significant(P>0.05);the secretion of E2 in serum of Turpan black sheep throughout the estrous cycle was:during estrus,late estrus Secretions of estrus,interestrus and preestrus are(8.106±1.203 pg/mL),(8.160±1.257 pg/mL),(8.134±1.392 pg/mL),(7.760 ±1.201 pg/mL),and pre-estrus The secretion amount was significantly lower than that of estrus,late estrus and interestrus(P<0.05).To sum up,the content of reproductive hormones in the serum of Turpan black sheep throughout the estrous cycle has obvious changes in each period or the entire cycle,and the content of each hormone is also significantly different.The estrous interval is mainly 16 to 17 days. |
