In this experiment,a layer of latex cover is coated on the surface of the sponge.This homemade progesterone plug is not easy to cause vaginal inflammation or bleeding.From the clinical trial results,it achieved good result of estrus synchronization and had certain practical value.This experiment adopted 24 h monitoring system to record individual behaviors of small-tail Han sheep ewes in fall.Daily behavior components were defined through preliminary observation.This paper makes an analysis and statistics to estrous cycle behaviors of small-tail Han sheep with scan sampling method to observe the main behaviors time distribution and the changes in different physiological period.These behavior data provide the basic data for breeding manager to improve management measure,meet the needs of small-tail Han sheep behavior and improve the welfare level.This paper aims to reveal the fecal hormones secretion regular of the small-tail Han sheep estrous cycle and explore a new kind of research approach for the research of sheep,cattle and other livestock hormone levels by detecting LH,FSH,E2,P4,Gn RH and PRL six hormones with ELISA.In addition,estrus induction were implemented in small-tail Han sheep by using progesterone vaginal plug,to analyze the effect of exogenous progesterone on the secretion levels of reproductive hormones,as well as to explore the correlation between daily behavior and hormone levels in small-tail Han sheep estrous cycle.The results are as follows:1.In the experiment of estrus synchronization by using homemade progesterone vagina plug in Hu-sheep,the estrus rate of lactating ewes was 85%,the estrus rate of non-pregnant ewes was 93.33%,the difference was not significant(p>0.05).The pregnancy rate of lactating and non-pregnancy ewes were 75% and 83.33%,the difference was not significant(p>0.05).These results suggest that progesterone vaginal suppository combined with intramuscular injection of PG have good effect on estrus synchronization in non-pregnancy ewes and lactating ewes.2.Small-tail Han sheep daily behavior including resting,ruminating,feeding,watching,exploratory,movement,grooming,defecation,urination and so on.Resting,ruminating and feeding three acts in turn account for 27.20% and 26.78% and 25.62% of day and night time.Watching,exploratory and moving behavior in turn account for 10.11% and 5.63% and 3.07%,Defecation and urination account for 0.10% of day and night time.3.Daily behaviors time allocation in Small-tail Han sheep diestrum and estrum is different.Resting behavior time allocation of M1 and M5 in estrum was significantly lower than that in diestrum(p<0.05).Movement and exploratory time allocation of M3 in estrum was significantly higher than that in diestrum and significant differences(P<0.05).Watching and movement time allocation of M5 in estrum was significantly higher than that in diestrum.The differences of daily behaviors time allocation of M6 and M7 between diestrus and estrus was not significant(p<0.05).The watching and movement Estrus time allocation of nulliparous sheep is higher than that of pluriparous ewes and the difference was significant(p<0.05).The above results show that the estrus performance of nulliparous ewe was more obvious than pluriparous ewe.It was related with sexual experience.4.The fecal P4,E2,LH,FSH,Gn RH and PRL average concentration of Small Tail Han sheep in turn were 1069.41pmol/L,786.41 pmol/L,1672.56 ng/L,10.01 IU / L,129.98 ng/L,519.90 ng/L.Among them,E2 and LH,PRL and Gn RH was correlated significantly(p<0.01),P4 and FSH was correlated significantly(p<0.05).The hormone levels measured in feces can accurately reflect the change of hormone levels of small-tail Han sheep.5.There are differences between small-tail Han sheep individuals on fecal hormone P4,E2 and Gn RH secretion.Compared with nulliparous sheep,the fecal Gn RH and P4 concentrations of pluriparous sheep was lower,which may be related to without significant performance in estrus.6.During the progesterone plug implantation,progesterone concentration gradually increased,Gn RH concentration showed a downward trend.The concentration of E2,LH,FSH and PRL increased rapidly after withdraw of plug,with intramuscular injection PG at the same time.E2 appeared multiple peaks,which mean that many follicular were raised to development.The low progesterone level after induction estrus may be associated with the intramuscular injection of PG after progesterone plug withdrawal.
