| Monogeneans belong to the Platyhelminthes Monogenea and an important aquatic organism.In this paper,the routine investigation and statistical methods of fish parasites investigate the introduction of some fish parasitic Monogenean in Xinjiang,the morphological and molecular biological techniques identify the species of Monogenean.The phylogeny of some worms was studied.At the same time,the distribution of parasitic fins in the host was statistically analyzed,and the types and infections of the introduced fish parasitic Monogenean were further clarified,and the pathogens were provided for the development of aquaculture in Xinjiang and the artificial introduction of fish in Xinjiang.Details are as follows:(1)Combine morphology and molecular biology methods to identify the Gyrodactylus of parasitic fishes on the surface of Salvinus fontinalis,Oncorhynchus masou masou,Oncorhynchus mykiss.The results show that the species of the Gyrodactylus brachymystacis,which will be obtained.The homology analysis of the ITS 1-5.8S-ITS2 gene sequence showed 99%(1078/1087)similarity to G.brachymystacis in GenBank,GC content of 44.7%,and 6 mutation sites.The phylogeny of Gyrodactylus brachymystacis was systematically studied to further clarify its taxonomic status.(2)Analysis of the infection rate and infection intensity of Gyrodactylus brachymystacis.The results show that the average infection rate of G.brachymystacis on Salfelinus fontinalis fins was 65.1 1%,and the tail fin infection was the highest G.brachymystacis.The highest intensity of infection on each fin is the caudal fin;the spatial distribution of G.brachymystacis on the fins of the host Salvelinus fontinalis is aggregated.It was found that the average infection rate of G.brachymystacis in Oncorhynchus masou masou was 89.98%,and the highest infection rate of G.brachymystacis was pectoral fin;while the highest infection intensity of G.brachymystacis on each fin was caudal fin;the spatial distribution of G.brachymystacis parasitic on the host Oncorhynchus masou masou is aggregated on each fin.(3)Seven species of Monogenean were identified by the traditional morphological identification methods,namely Onchocleidus dispar,Cichlidogyrus sclerosus,Cichlidogyrus sp,Gonocleithrum aruanae,Gonocleithrum coenoideum,Gussevia asota,Ligictaluridus pricei,among which O.dispar,G.aruanae,G.coenoideum is a new record species in China.This paper compares O.dispar with similar species and plots G.aruanae and G.coenoideum.(4)The 28S rDNA sequences of five Ancyrocephalinae were sequenced by conventional molecular biology techniques,and one gene accession number(MG596661)was obtained from NCBI.The remaining sequences were to be uploaded,and combined with the Ancyrocephalinae worm downloaded from GenBank.The 28S rDNA sequence,which maps phylogenetic trees,indicates the location of these worms in phylogeny and further clarifies their taxonomic status. |
