The egg production performance of geese is an important economic trait in production.Enhancing the egg production ability of geese can not only promote the breeding of geese,but also promote the development of the geese industry and increase production efficiency.The egg production ability of geese belongs to low heritability traits.The development of effective molecular markers for molecular breeding can rapidly increase the egg production of geese in production.SNP Record-106582(rs 1714766364)is a mutation site that was screened by our laboratory through simplified genome sequencing.After sequence comparison,this SNP is located in the promoter region of the ACSF2 gene.Although it has been demonstrated in the fifth and seventh generations that SNPs are significantly associated with egg production,the specific mechanism of SNP is still unclear.In order to study the effect of SNP on egg production performance,this study intends to investigate the effects of SNPs on the function of the mapped genes and explore the specific mechanisms of action of SNPs.The results have been presented as follows:Compared with different genotypes of SNP Record-106582,we found that the egg production performance of AA genotypes was better than that of CC genotypes(P<0.05),there was no significant difference between AC genotypes and other genotypes.In addition,the expression of ACSF2 mRNA in AA genotype was significantly lower than that in CC genotype(P<0.05)(n=12),and was confirmed by detecting the transcriptional activity of the promoter of different genotypes(Activity of AA genotypes was significantly lower than that of CC type).Overexpression of ACSF2 gene in follicular granulosa cells of Yangzhou Goose,we detected significant changes in the expression of some genes involved in energy metabolism and the concentration of ATP.The results showed that the concentration of ATP increased significantly(P<0.0l)after overexpression of ACSF2 gene.It is speculated that the ACSF2 gene influences the concentration of ATP by regulating the energy metabolism in granule cells,leading to the apoptosis of granulosa cells(the increase of ATP concentration activates a cascade of apoptosis).SNPs in the promoter region lead to significant differences in transcriptional activity,generally due to different transcription factor binding at this SNP site.We used the website to predict differences of SNP-binding transcription factors.The result show that AA promoter region of the ACSF2 gene binds to the transcription factor c-myb,when the A mutation is C,the promoter region can’t bind to c-myb.We detected the transcriptional activity of three reporter luciferase reporter vectors of different lengths of the ACSF2 gene promoter region and found that there was a silencer between-430bp and-887bp(there was a negative regulatory transcription factor binding site between-430 and-887).C-myb can be used as a negative transcription factor.We speculated that the ACSF2 gene inhibits ACSF2 gene expression by binding to the transcription factor c-myb,reducing ATP levels,and ultimately increasing the egg production of Yangzhou geese.We identified the specific molecular mechanism of this SNP in the ACSF2 gene leading to differences of egg production,provided an effective and viable option for breeding geese.
