| Objective This article aims to study the expression of AIRE,NFKBIA and adhesion factors ICAM-1,VCAM-1 and LAMA-1 in the thymus of TAMG patients.In addition,we also explored their relevance to the pathogenesis of MG.Methods1.For the detection of RNA transcription level,select TAMG patients(MGT group)and patients with simple thymoma(ST group)who underwent thymoma resection in the Thoracic Surgery Department of Jilin University First Hospital from2017 to 2020,and collect thymoma specimens.At the same time,patients in the first hospital of Jilin University who removed part of the thymus to expose the pericardium during cardiac surgery were selected as the HC group.Patients with acute infections were excluded.The above-mentioned patients all signed the informed consent form and passed the review of the ethics committee.2.Quantitative RT-PCR technology was used to detect the m RNA transcription levels of AIRE,CHRNA1,ICAM-1,VCAM-1,LAMA-1,NFKBIA in the thymus of the HC group and the other two groups of thymoma tissues.Western blotting was used to quantitatively detect the protein levels of AIRE,CHRNA1,ICAM-1,and VCAM-1.Results1)Quantitative RT-PCR1.There was no statistically significant difference in AIRE m RNA transcription levels between MGT group and ST group,and between HC group and ST group(p>0.05).The transcription level of AIRE m RNA in the MGT group was significantly lower than that in the HC group,and the difference was statistically significant(p=0.014).2.There were no statistically significant differences in NFKBIA m RNA transcription levels between MGT group and ST group,and between MGT group and HC group(p>0.05).The transcription level of NFKBIA m RNA in ST group was significantly higher than that in HC group,and the difference was statistically significant(p=0.001).3.There was no statistically significant difference in ICAM-1 m RNA transcription levels between MGT group and ST group,and between MGT group and HC group(p>0.05).The transcription level of ICAM-1 m RNA in ST group was significantly higher than that in HC group,and the difference was statistically significant(p=0.005).4.The transcription level of VCAM-1 m RNA in ST group was significantly higher than that in MGT group(p=0.007)and HC group(p=0.015),and the difference was statistically significant.There was no statistically significant difference in VCAM-1 m RNA transcription levels between the MGT group and the HC group(p=1.000).5.Comparing the three groups,there was no significant difference in the transcription levels of CHRNA1 m RNA and LAMA-1 m RNA among the three groups(p>0.05).2)Western Blotting1.There was no statistically significant difference in AIRE protein expression between MGT group and ST group,and between HC group and ST group(p>0.05).The expression level of AIRE protein in the MGT group was significantly lower than that in the HC group,and the difference was statistically significant(p=0.049).2.The expression level of NFKBIA protein in ST group was significantly higher than that in MGT group(p=0.027)and HC group(p=0.022),and the difference was statistically significant.There was no statistically significant difference in NFKBIA protein expression between the MGT group and the HC group(p=1.00).3.The expression level of ICAM-1 protein in HC group was significantly lower than that in ST group(p=0.009)and MGT group(p=0.017),and the difference was statistically significant.There was no statistically significant difference in ICAM-1protein expression between MGT group and ST group(p=1.00).4.There was no significant difference in the expression level of VCAM-1protein among the three groups(p=0.193).Conclusion1.The transcription and expression of AIRE protein in the thymus tissue of TAMG patients are significantly reduced,suggesting that low AIRE may be related to the pathogenesis of MG.2.There is no significant difference in the transcription level of CHRNA1 in the thymus tissue of patients with simple thymoma,TAMG patients and healthy patients.3.The low expression of NFKBIA protein in TAMG group may lead to weakening of NF-κB inhibition,making inflammation and immune disorders in the thymus microenvironment more likely to occur,and more likely to cause MG.4.The protein expression of ICAM-1 in the thymus tissue of TAMG patients is increased.However,under the background of low expression of AIRE and low expression of NFKBIA,the increased affinity between cells may be more helpful to trigger abnormal immune responses.5.LAMA-1 found no meaningful results in this study. |
