Objective To investigate whether lactoferrin(LF)can alleviate acute radiation intestinal injury caused by radiation in mice and its mechanism,and to provide a theoretical basis for the research strategy of using lactoferrin as a protective agent for radiation intestinal injury.Methods Fifty male C57BL/6J mice were randomly divided into normal control group,single irradiation group,2mg LF+ irradiation group,4mg LF+ irradiation group,and 6mg LF+ irradiation group,with 10 mice in each group.A single irradiation of14 Gy X-ray was given to four groups of mice on Varian 23 Ex accelerator,and the irradiation range was as follows: Mice abdomen below xiphoid process,pubic symphysis above,among them 2 mg LF + exposure group,4 mg LF + exposure group,6 mg LF + light begin 7 days before irradiation and irradiation in mice after 3 days to fill the stomach to LF,observe the general situation and gastrointestinal reaction of mice,3 days after exposure to death mice,small intestinal tissue in mice,using hematoxylin-eosin(HE)staining to observe each group of mice small intestine mucosa pathological morphological changes and damage degree,and ELISA method in mice serum Diamine Oxidase(DAO),the level of Transforming growth factor-β(TGF-β)testing mice intestine tissue homogenate Malondialdehyde(MDA),Superoxide Dismutase(SOD)and Nitric Oxide(NO)levels.Results Compared with the normal control group,the body weight of mice in the simple irradiation group was significantly reduced.Some intestinal villi were broken off,some epithelial structures disappeared,crypt arrangement was disorderly(some were lost),and the glandular cavity was infiltrated by inflammatory cells.The levels of DAO and TGF-β in serum were significantly increased(P<0.05),the contents of MDA and NO in small intestine were significantly increased(P<0.05),and the activity of SOD was significantly decreased(P<0.05).Compared with the irradiation group,the pathological rupture of intestinal villi and loss of intestinal mucosa of mice in 2mg LF+ irradiation group were alleviated,the serum DAO level decreased(P<0.05),the serum TGF-β level and the contents of MDA,NO and SOD in small intestine were not significantly changed(P>0.05).4 mg LF + exposure group and 6 mg LF + exposure group mice intestine mucosa pathological morphology(intestinal villus and fossae)relatively simple exposure group gradually improving,among them 6 mg LF + exposure group villi structure basic returned to normal,neat fossae,serum DAO,TGF-beta levels decreased(P < 0.05),decreased MDA and NO content in intestinal tissues(P < 0.05),SOD activity increased(P < 0.05).Conclusion Lactoferrin can reduce the oxidative stress response in the small intestine of mice,reduce the secretion of inflammatory factors,inflammatory response,and the degree of intestinal mucosa injury,and has a certain protective effect on the acute radiation intestinal injury of mice.
