Expression And Significance Of Claudin-8 In Clear Cell Renal Cell Carcinoma

Objective:Claudin protein family is known to be closely related to the occurrence、development and metastasis of many malignant tumors.The aim of this study was to investigate the expression of Claudin-8 in clear cell renal cell carcinoma(ccRCC)and adjacent tissues,and to explore the clinical and pathological parametes related to Claudin-8 in clear cell renal cell carcinoma,and the correlation with the survival time of the patients after operation.Methods:After radical nephrectomy in the Department of Urology of Liaoning Provincial Cancer Hospital,63 cases(16 cases of Grade I,37 cases of Grade II,10 cases of Grade Ⅲ)and 12 cases of adjacent tissue were confirmed as ccRCC by pathology.The expression of Claudin-8 in ccRCC and adjacent tissuses was confirmed by two-step immunohistochemical.The relationship between Claudin-8 and clinical pathological parameters and prognosis of patients with ccRCC was analyzed by statistical software.Results:There was significant difference in the expression of Claudin-8 between ccRCC and adjacent tissue(P<0.05).The high expression of Claudin-8 was 25.4%in ccRCC and 83.3%in adjacent tissue.The expression of Claudin-8 in ccRCC was significantly lower than that in adjacent tissue.The down-regulation of Claudin-8 expression was associated with the clinical stage and tumor size of ccRCC(P<0.05).In contrast,the expression of Claudin-8 had no significant difference with age,sex,lymph node metastasis,T stage and pathological grade of patients with ccRCC(P>0.05).Using Kaplan-Meier method and Log-Rank test,the relationship between the survival time of patients with ccRCC and the expression of Claudin-8 was analyzed.The results showed that the 5-year survial time of low-expression Claudin-8 group was significantly lower than that of high-expression Claudin-8 group,there was significant difference betweeen them(P=0.026,P<0.05).Conclusions:The results of this study showed that the expression of Claudin-8 was down-regulated in ccRCC patients compared with the adjecent tissue,and correlated with the clinical stage and tumor size of patients with ccRCC,and influenced the survival time of patients with ccRCC.
