The Research Of Quality Standard And Concentarted Herb Granule Of Boehmeriae Rhizoma Et Radix

Boehmeriae rhizoma et radix belongs to Boehmeria nivea(L.)Gaud dry roots and rhizomes.The roots and rhizomes has been commonly used in the treatment of heat,hemostasis,tocolysis,detoxification.Currently,the basic research related to Boehmeriae rhizoma et radix is still very limited.There is no effective quality standards,seriously restricting the promotion and application of the herbs.Concentrated Herb Granule is a single herbal medicine processing from the extraction.It has the advantages of small size,easy to carry and store,stable and controllable quality and so on.The purpose of this paper is to establish a improved quality and preparation standard,lay a material foundation for controlling quality of Boehmeriae rhizoma et radix and explore the preparation process and quality evaluation of its granules.1)Study on standard of medicinal materials.The powder characteristics of Boehmeriae rhizoma et radix were studied,and the main microscopic characteristics of Boehmeriae rhizoma et radix were described.Thin layer chromatography(TLC)method was established for the identification of Boehmeriae rhizoma et radix.Medicinal powder was heated under reflux in ethanol,and the filtrate was concentrated.By treatment of neutral alumina column,70% ethanol elution liquid was concentrated and pointed to silica gel G thin layer plate,developing system chloroform-methanol-cyclohexane(7:1:5)was applied.Then it was sprayed with ferric chloride testing solution,and heated to the spot color clear in the 105℃.The same color spots in the TLC graphs of sample existed at the corresponding position compared with the controls.According to the inspection items of “China pharmacopoeia” four general principles(2015 edition)and alcohol-soluble extraction of Boehmeriae rhizoma et radix were determined.Moisture content is not more than 13.0%.Total ash content is not more than 8.5%.Acid-insoluble ash content is not more than 2.0%.The standard of medicinal materials of ramie root was established,and was included in the 2016 edition of "standard of Chinese herbal medicine in Hebei province".2)The HPLC fingerprint method for quality control of Boehmeriae rhizoma et radix and Pteridophyta was established,and methodolody was also researched.The precision and repeatability of this method were well conformed to the standard.This study provided scientific basis for the quality control of Boehmeriae rhizoma et radix.3)Study on concentrated herb granule.Based on the standard decoction,the extraction,concentration,drying and granulation technology of Boehmeriae rhizoma et radix extracts were investigated.The preparation process of Boehmeriae rhizoma et radix extracts was determined by using the paste rate.It was as follows as soaking seawater 0.5h,adding 10 and 8 folds water,extracting for 2 times,lasting for 1h and 0.5h.Concentrated method for the concentration is decompression;drying method is spray drying,adding dextrin amount of 3%;granulation method is dry granulation.Pilot test was carried out to verify the feasibility of the test.The optimum results were compared with the standard decoction of evaluation which was to verify the feasibility of process optimization.By examining the samples of the pilot sample,the limits of water and extract were determined.The differences of particle size,solubility and loading were investigated.The quality standard of Boehmeriae rhizoma et radix extracts was established.
